Expression Analysis of CaWRKY Fused Proteins

To test the expression level of fused protein, A. tumefaciens GV3101 cells harbouring CaWRKY40‐HA, CaWRKY27‐HA, or CaWRKY58‐HA were coinfiltrated with pCaWRKY40::GUS into pepper leaves. Infiltrated plants were grown in the greenhouse for another 2 days. The total protein of the infiltrated leaves was extracted using protein extraction buffer (10% glycerol, 25 mM Tris‐HCl, pH 7.5, 150 mM NaCl, 1 mM EDTA, 2% Triton X‐100, 10 mM DTT, 1 × complete protease inhibitor cocktail [Sigma‐Aldrich], and 2% [wt/vol] polyvinylpolypyrrolidone [Choi et al., 2012]). Immunoprecipitation of HA-tagged Proteins

Total protein extracts were incubated with anti-HA agarose (Thermo Fisher Scientific, Waltham, MA, USA) overnight at 4 °C.
