Cck 8

Manufactured by BBI Lifesciences
Sourced in China

CCK-8 is a colorimetric assay kit used to measure cell viability and cytotoxicity. It utilizes the water-soluble tetrazolium salt WST-8 to produce a water-soluble formazan dye upon reduction in the presence of an electron carrier. The amount of the formazan dye generated is directly proportional to the number of living cells.

Cell clone formation assay: The cells were transfected in 6-well plates (500 cells per well) and fixed with 4% paraformaldehyde. The cells were stained with crystal violet, and the number of clones was counted as >50 cells/group.
CCK-8 assay: After transfection and other treatments in a 96-well plate (about 1,500 cells per well, 3 parallel wells), a volume of 10 μl of CCK-8 (BBI Life Sciences, China) was mixed with 90 μl of culture medium at 0, 24, 48, 72, and 96 h after transfection, and incubated for an additional 2 h at 37°C. A microplate spectrophotometer (Biotek, USA) was utilized, and the absorbance was measured at 450 nm.

Cell Proliferation Assay Protocol

Check if the same lab product or an alternative is used in the 5 most similar protocols
The cells (BCPAP and TPC-1) were plated in 96-well plates at 200 µl per well and incubated with DMEM containing 10% FBS at 37°C. CCK-8 (BBI Life Sciences Corporation) was used to determine the proliferation of cells at various time points (0, 24, 48 and 72 h). The cells were incubated with 10 µl CCK-8 solution for 1 h at 37°C, and the absorbance at 450 nm was then measured (Biotek EPOCH2, BioTek Instruments, Inc.). Each experiment consisted of five samples. Every independent experiment was performed in triplicate.

Cell Viability Assay Using CCK-8

Check if the same lab product or an alternative is used in the 5 most similar protocols
Neuronal viability was assessed with a cell viability counting kit CCK-8 purchased from BBI Life Sciences (Shanghai, China). Briefly, cells after treatment were collected following the manufacturer’s protocol. Cell suspension at 100 μl was transferred to a 96-well plate at a final concentration of 500,000 cells/ml. Cells were incubated with 10 μl CCK-8 solution from the kit per well for 4 h and assessed with a PerkinElmer spectrometer (PerkinElmer, USA) at the absorbance of 450 nm.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
