Serum Biomarkers for Bone Turnover

Serum samples were collected at the time of sacrifice and stored at −80°C. Serum levels of Pro-collagen type 1 N terminal propeptide (P1NP) were measured using the Rat/Mouse P1NP EIA kit (Immunodiagnostic Systems, Fountain Hills, AZ, USA) and serum C-terminal telopeptide of type I collagen (CTX-1) levels were measured using RatLaps (CTX-1) EIA kit (Immunodiagnostic Systems Inc., Gaithersburg, MD, USA). The MILLIPLEX MAP mouse angiogenesis magnetic bead panel – multiplex assay was used to measure serum VEGF-A levels (MAGPMAG-24K, EMD Millipore, Billerica, MA, USA). All the assays were performed as recommended by the manufacturers and all the samples were assayed in duplicate. A standard curve was generated from each kit, and the absolute concentrations were extrapolated from the standard curves. The coefficients of variations for inter-assay and intra-assay measurements were < 10% for all assays and are similar to the manufacturers’ references [28 (link),29 (link)].

Measuring Serum CTX-I Levels

Check if the same lab product or an alternative is used in the 5 most similar protocols
Serum samples were collected at the time of euthanization and stored at −80 °C. Serum C-terminal telopeptide of type I collagen (CTX-I) levels, a biomarker of bone resorption, were measured using RatLaps (CTX-1) EIA kit (Immunodiagnostic Systems Inc., Gaithersburg, MD, USA). the assays were performed follow the manufacturer’s protocol and all the samples were accessed in duplicate. The absolute concentrations were extrapolated from the standard curves which generated from the kit.

Serum Biomarkers of Bone and Inflammation

Check if the same lab product or an alternative is used in the 5 most similar protocols
P1NP levels in mouse serum were determined using the Rat/Mouse P1NP EIA kit (Immunodiagnostic Systems). C-terminal telopeptide of type I collagen (CTX-I) levels in fasting mouse serum were determined using the RatLaps CTX-I EIA kit (Immunodiagnostic Systems). Serum cytokine arrays were performed using 44-Plex technology (MD44) from Eve Technologies. IL-20, IL-11, Lif, and IL-1β levels were determined using mouse IL-20 (EMIL20; ThermoFisher), IL-11 (EMIL11; ThermoFisher), Lif (MLF00; ThermoFisher), and IL-1β ELISA kits (5018265; ThermoFisher). Absorbance was measured at 450 nm using Clariostar Plate Reader or Molecular Devices FilterMax.

ELISA was performed on serum samples. CTX was measured using the RatLaps CTX-I EIA kit (Immunodiagnostic Systems, Tyne & Wear, UK), OIM serum was diluted up to 1:8 to ensure values fell within the standard curve. Cytokines were measured with Mouse TNFα Quantikine High Sensitivity ELISA kit and Mouse IL1α Quantikine ELISA kit (R&D Systems). All assays were performed according to the manufacturer’s instructions.

Calvarial Bone Resorption Assay

Check if the same lab product or an alternative is used in the 5 most similar protocols
Calvariae from 5-d-old wild-type or DKO mice were isolated aseptically, cleaned, and cultured for 16 h at 37°C in 0.5 ml of BGJb medium (Life Technologies) containing 1 mg/ml BSA (fraction V; Sigma-Aldrich; Moxon et al., 2015 (link)). Half calvariae were transferred to fresh medium with 0.1 μM parathyroid hormone (H-4835.0005; Bachem) in the presence or absence of an anti–galectin-3 monoclonal antibody (sc-32790L; Santa Cruz) or an anti-Lrp1 monoclonal antibody (MA1-27198; Thermo Fisher Scientific), and cultured for an additional 5 d. Culture supernatant was collected for bone resorption marker CTX-I level detection using RatLaps CTX-I EIA kit (AC-06F1; Immunodiagnostic Systems). The half calvariae were either fixed for immunostaining with an anti-TRAP polyclonal antibody (sc-30833; Santa Cruz), an anti–galectin-3 monoclonal antibody (#125401; Biolegend; clone M3/38), or an anti-V-type proton pump-3 (Vpp3) polyclonal antibody (ab200839; Abcam) as describe above or snap-frozen for TRAP activity assay.

Serum Osteocalcin and CTX Measurement

Check if the same lab product or an alternative is used in the 5 most similar protocols
Circulating osteocalcin and CTX in serum was measured using a mouse Osteocalcin enzyme immunoassay kit (Thermo Fisher) and RatLaps (CTX-I) EIA kit (Immunodiagnostic Systems, Boldon, UK) respectively according to the manual provided by manufacturers. Blood was collected by retro-orbital bleeding into 1.7 mL microcentrifuge tubes. Blood was then kept at room temperature for one hour and centrifuged at 1500 x g for 10 min to separate serum from cells.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
