The pEGFP(C1)-PP1α and pEGFP(C1)-PP1γ plasmids were previously described14 (link) and are available through Addgene (plasmids #44224 and #44225). The C-terminal truncation mutant pEGFP(C1)-PP1γΔ299–323 was derived from the full-length construct by PCR-based insertion of a premature STOP codon. The C-terminal chimera pEGFP(C1)-PP1γ(1–298)PP1α(299–330) was derived by PCR-based insertion of a SalI restriction site into the full-length construct at the relevant site, followed by digestion and ligation of the PP1α C-terminal coding sequence. The pEGFP(C1)-RRP1B and pEGFP(C1)-RRP1BRAxA (V684A/F686A) plasmids were previously described.15 (link) PP1 and RRP1B point mutants were derived using Quickhange mutagenesis (Agilent Technologies) and sequence confirmed. Antibodies that recognize RRP1B, RepoMan and ASPP2 were obtained from Abcam, PP1g antibodies were from Santa Cruz and GFP-antibodies from proteintech®. HRP-conjugated secondary antibodies were from ThermoFisher.

Comprehensive Western Blot Antibody Panel

The following antibodies were used: FIH-1, Grb2, tubulin, GAPDH, Notch1, GADD45α, FLAG (Santa Cruz Biotechnology, Inc., Santa Cruz, CA, USA), p63α, E-cad, STAT1, pY701-STAT1, Plectin1, HDAC1, (Cell Signaling Technology, Danvers, MA, USA), ASPP2 (Abcam, Cambridge, United Kingdom), and β-catenin (Sigma-Aldrich Corp., St Louis, MO, USA).
