The molecular weight (Mw) of EPSKar1 was determined by size-exclusion chromatography using Agilent 1260 Infinity Multi-Detector GPC/SEC System (Agilent 1260 Infinity Series, USA) with refractive index detector and PL aquagel column (Agilent Technologies). Briefly, 2 mg of EPSKar1 was dissolved in 1 mL distilled water and filtered through a 0.22-μm pore size filter. Data collection and processing were performed using the Agilent GPC/SEC MDS software. The corresponding molecular weight of EPSKar1 was determined using a standard curve of dextran with different molecular weights (5.20, 11.60, 23.80, 48.60, 148.00, 273.00, 410.00, and 668.00 kDa).

Analyses were performed with buffer citrate/phosphate 0.15 M + 0.1 M NaCl pH 5 as eluent with a flow of 1 mL/min and poly(ethylene oxide) standard (40 kDa) to obtain the calibration curve. The column temperature was set at 30 °C.
