Cells were grown on high-precision borosilicate cover glasses (LH22.1 Roth Labware). For cyclin B staining (1:200; Cell Signalling), cells were fixed in PTEMF buffer or in 4% PFA followed by ice-cold methanol, and stained as previously described [29 (link)]. For γH2AX and phospho-RPA staining, cells were fixed in 4% PFA, permeabilised in 0.5% Triton X-100 in PBS and blocked for one hour in 0.1% Tween and 1% BSA, followed by incubation with primary antibodies: rabbit anti-γH2AX (1:500; Bethyl) or rabbit anti-phospho-RPA S4/S8 (1:500; Bethyl) and subsequent incubation with appropriate secondary antibodies (1:500; Alexa Fluor®).
