Steroid and Phthalate Standards Characterization

Four steroid standards including E1, E2, E3 and EE2 and BPA were supplied by Sigma-Aldrich (USA). The standards of six phthalates, including DMP, DEP, DBP, BBP, DEHP and DnOP were obtained from Aladdin Reagent Corporation (China). HPLC grade methanol, alkane, acetone, methyl tert-butyl ether (MTBE) and ethyl acetate were supplied by CNW (CNW Technologies GmbH, Germany). Dimethyl sulphoxide (DMSO), sodium hydroxide (NaOH) and a 25% ammonia solution were supplied by Merck (Darmstadt, Germany). Dulbecco's Modified Eagle Medium (DMEM without phenol red (Gibco)), sodium pyruvate (100 mM, sterile-filtered), alpha-Minimal Essential Medium (α-MEM (Gibco)), penicillin-streptomycin, fetal bovine serum (FBS), charcoal-stripped FBS, L-glutamine (200 mM), trypsine (0.5% (Gibco)), phosphate buffered saline (PBS, 1X, pH 7.4) and trypsine without phenol red (10X, 0.5% (Gibco)) were purchased from Life Technologies (United Kingdom). Ultrapure water was obtained using a Milli-Q Advantage A-10 system (Millipore, USA). The physiochemical properties of the investigated steroids, BPA and phthalates are provided in Table S1.

Dexamethasone Injection Quantification

BPA Quantification by LC-MS/MS

The BPA standard was purchased from Aldrich-Sigma (purity >98%, St. Louis, MO, USA). BPA-13 C12 (Cambridge Isotope Laboratories Ins, USA), HPLC-grade methanol, ethyl acetate and ammonium acetate (CNW Technologies) were purchased from Shanghai ANPEL Scientific Instrument Co., Ltd. β-Glucuronidase from Helix pomatia (Type HP-2, aqueous solution, β-glucuronidase activity ≥100000 units/mL, sulfatase activity ≤7500 units/mL) was purchased from Sigma-Aldrich. Deionized water for the preparation of buffers was purified by a Milli-Q ® water-purification system (Millipore, Bedford, MA, USA). An Oasis HLB cartridge (Waters, 200 mg/6 cm 3 , Milford, MA, USA) was purchased from Waters.
Stock solutions of BPA and 13 C12-BPA were prepared at a concentration of 1 mg/mL in methanol. All stock solutions were stored at -20°C. Calibration standards, ranging from 0.20 to 25 μg/L, were prepared from the stock solution through serial dilution by using a methanol-water mixture (50/50, v/v); isotope stock solutions ( 13 C12-BPA, 100 μg/L) were also prepared by using the same methanol-water mixture.

Plasma Metabolite Extraction and Derivatization

An aliquot of 50 µL plasma was mixed with 200 µL of methanol (HPLC grade; CNW Technologies, Duesseldorf, Germany) containing 25 µmol/L 2-chloro-L-phenylalanine (Shanghai Hengbai Biotech, Shanghai, China) as an internal standard, which was used for normalization. After vortexing for 30 s, samples were ultra-sonicated in iced water for 10 min. After centrifugation at 12 000 rpm at 4 °C for 15 min, 180 µL of supernatant for each sample were transferred into a new tube and dried in a vacuum concentrator. The residues of each sample were then added with 20 µL of 20 mg/mL methoxyamine hydrochloride (AR grade; TCI, OR, USA) in pyridine (HPLC grade; Adamas Pharmaceuticals, CA, USA). The above samples were kept at 80 °C for 30 min, after which 40 µL of bis-(trimethylsilyl) trifluoroacetamide (BSTFA) in 1% TMCS (REGIS Technologies, IL, USA) was added and samples were incubated at 70 °C for 1.5 h. The samples were stored at room temperature until analysis. Quality control (QC) samples were prepared by pooling an aliquot of 30 µL from each sample.
