Rat Model of Subarachnoid Hemorrhage: Evaluating Neuroprotective Drugs

Sixty rats were randomly divided into 5 groups (12 rats in each group):
Group A: Sham operation group (n = 12) was divided into three groups: 2nd, 4th and 6th days, injecting saline into subarachnoid space.
Group B: Blood injection model group (n = 12) was divided into three groups 2nd, 4th and 6th days, injecting autologous blood into subarachnoid space;
Group C: Nimodipine (Sigma Company, USA) group (n = 12) was divided into three groups: 2nd, 4th and 6th days. Three groups were given Nimodipine orally (2 mg/kg/d) on the premise of blood injection;
Group D: Flunarizine hydrochloride (Sigma Company, USA) group (n = 12) was divided into three groups: 2nd, 4th and 6th days. Three groups were given Flunarizine hydrochloride (1 mg/kg/d) by intragastric administration on the premise of blood injection;
Group E: Normal group (n = 12) was divided into 3 groups: 2nd, 4th and 6th days without any treatment.

Screening Cytotoxic Compounds in EP1/GFP Cells

1 mL cultures of EP1/GFP reporter cells were plated at 100,000 cells/ml and treated with 33 μM of eflornithine (Sigma-Aldrich 1234249), phenothiazine (Sigma-Aldrich P14831), spironolactone (Sigma-Aldrich S3378), or vehicle control and counted daily by hemocytometer. Each day following counting, cells were diluted back to 100,000 cells/ml. For growth curve analysis by flow cytometry, 200 μl cultures of parasites were plated at 100,000 cells/ml and treated with 33 μM of phenothiazine (Sigma-Aldrich P14831), triprolidine hydrochloride (Sigma-Aldrich T6764), flunarizine hydrochloride (Sigma-Aldrich F8257), aprepitant (Sigma-Aldrich SML2215), bufexamac (Sigma-Aldrich B0760), clemastine fumarate salt (Sigma-Aldrich SML0445) or vehicle control. Parasites were stained daily with Sytox Orange and a fixed volume of cells was analyzed by flow cytometry. The number of live cells was calculated as the number of Sytox Orange negative cells falling within the live gate. Each day following flow analysis, cells were diluted back to 100,000 cells/ml according to the number of live cells counted.

Screening FDA Pharmakon Library for Anthelmintic Compounds

The FDA Pharmakon compound library was purchased from MicroSource Discovery Systems, Inc. (USA). Compounds were delivered in microplates (10 mM, dissolved in DMSO) and kept at -80°C until use. For in vivo studies, flunarizine hydrochloride, pimozide, nicardipine hydrochloride, oxethazaine, menadione, clofazimine, doramectin and metitepine mesylate were purchased from Sigma-Aldrich (Buchs, Switzerland) and fendiline hydrochloride, manidipine hydrochloride and lomerizine hydrochloride were purchased from Santa-Cruz Biotechnology (California, USA). Hanks Balanced Salt Solution (HBSS) was obtained from Gibco (Lucerne, Switzerland). Culture medium components for NTS and adult worms were obtained as follows: Medium 199 RPMI 1640 and penicillin (100 U/ml) and streptomycin (100 μg/ml) were purchased from Lubioscience (Lucerne, Switzerland) whereas inactivated fetal calf serum (iFCS) was purchased from Connectorate AG (Dietikon, Switzerland).
