Radioactive Serotonin Binding Assay

Tritium-labeled Serotonin creatinine sulfate (3H-serotonin; 28.3 Ci mmol−1 and 41.3 Ci mmol−1) and radiocarbon-labeled serotonin binoxalate (14C-serotonin; 54.0 mCi mmol−1) were obtained from Perkin Elmer (Waltham, MA, USA). Serotonin creatinine sulfate, imipramine hydrochloride, citalopram hydrobromide, 3,4-methylenedioxymethamphetamine hydrochloride (MDMA, “Ecstasy”), L-ascorbic acid, and Hanks’ Balanced Salts Solution (HBSS) were obtained from Sigma-Aldrich (St. Louis, MO, USA). Pargyline hydrochloride was from Cayman Chemical (Ann Arbor, MI, USA). Other drugs were generous gifts from manufacturers.

Investigating MDMA-Induced Sensory Gating Deficits

All drugs were diluted and prepared to an injection volume of 1 mL/kg on the day of the testing session. Drug doses used were in accordance with previous literature. A 2 mg/kg dose of MDMA ((±) 3,4 methylene-dioxymethamphetamine hydrochloride, Sigma Aldrich, St. Louis, MO, USA) was injected intraperitoneally. This was previously shown to elicit a paired-pulse sensory gating deficit [25 (link)]. Pretreatment drugs, haloperidol (Serenace®, Aspen Pharma Pty Ltd, St Leonards, NSW, Australia) 0.25 mg/kg, SCH23390 (Tocris Bioscience, Bristol, UK) 0.1 mg/kg, ketanserin tartrate (Tocris Bioscience, Bristol, UK) 2 mg/kg and WAY100635 maleate (Tocris Bioscience, Bristol, UK) 1 mg/kg, were all injected subcutaneously. The doses of these drugs were chosen on the basis of literature and preliminary experiments. Saline was injected subcutaneously for pretreatment control, but intraperitoneally for the treatment control injection. The pretreatment drug (saline or antagonist) was administered 30 min prior to the treatment drug (saline or MDMA). Recordings began 10 min after the treatment injection to allow time for the drug to take its effects and for the rat to settle into the chamber.

Synthesis and Characterization of Novel Psychoactive Substances

Ethylene glycol dimethacrylate (EGDMA), divinylbenzene (DVB), trimethylolpropane trimethacrylate (TRIM) and methacrylic acid (MAA) were purchased from Acros (Loughborough, UK). Prior to polymerization, the stabilizers in the monomers and crosslinker monomers were removed by passing the solutions over a column packed with alumina. Initiator 4,4′-azobis(4-cyanovaleric acid), ketamine hydrochloride, 3,4-methylenedioxymethamphetamine hydrochloride (MDMA), caffeine and solvents were obtained from Sigma Aldrich (Gillingham, UK). Diphenidine and the positional isomers of methoxphenidine (Scheme 1) were prepared, under UK Home Office license, as their corresponding hydrochloride salts using the procedure reported by Geyer et al. 5 The street sample was obtained from BRC Fine Chemicals (http://www.brc-finechemicals.com) prior to the legislative change. The Psychoactive Substances Act (2016), which came in effect on the 26 th May 2016, makes it an offence to manufacture, supply, possess and import and export psychoactive substances named in this Act, which includes MXP derivatives and their isomers. Structure and purity (>99%) of the target compounds was determined by NMR analysis and GCMS respectively. 5 All solutions were prepared with deionized water of a resistivity of 18.2 Ω cm or with home-made phosphate buffered saline (1× PBS) solutions.
