Nanoparticle-induced Cardiac ATP Depletion

ATP was measured using the Cell Titer Glow kit (Promega) in isolated ventricle myocytes (104 cells) after 24 h exposure to nanoSiO2 at LD50, and in ventricle tissue after perfusion of nanoSiO2 in the isolated heart. Cells or N2 frozen-grinded tissue in p96 well format were lysed according to the manufacturer instruction to record the luminescence derived from the ATP-dependent luciferase activity, using a microplate luminometer. ATP was quantified using a standard curve and normalized to the total protein in the sample.

Cellular ATP and Lactate Assay
