Serum Liver Enzyme Evaluation in Rat Model

Check if the same lab product or an alternative is used in the 5 most similar protocols
After contrast-enhanced ultrasonography at the indicated time points (normal group, 1- to 5-week groups after modeling) (Figure 2), 2 mL blood was drawn from the medial canthus vein of each rat, and the serum was separated and stored at −80 ℃ for later use. The level of serum alanine aminotransferase (ALT), aspartate aminotransferase (AST) and alkaline phosphatase (ALP) clinically used to evaluate hepatic aGVHD was detected by a Mindray® automated hematology analyzer (BS-350E, Shenzhen Mindray Bio-Medical Electronics Co., Ltd, Shenzhen, China).

Serum Lipid Profile Analysis in Mice

Check if the same lab product or an alternative is used in the 5 most similar protocols
After 12 h of fasting, mice were anesthetized, and blood samples were collected via cardiac puncture. Serum lipid profiles, including total cholesterol (TC), total triglyceride (TG), low‐density lipoprotein cholesterol (LDL‐C), and high‐density lipoprotein cholesterol (HDL‐C) were measured by enzymatic assay with an automatic biochemical analyzer (Mindray, BS‐350E), respectively.

Serum Biochemical Analysis in Mice

Check if the same lab product or an alternative is used in the 5 most similar protocols
Mouse peripheral blood was collected and centrifuged to obtain serum for analysis. We used a Mindray automated biochemical analyser (BS-350E) to test the serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), TG and total cholesterol (TC) levels.

Serum Protein Analysis in Mice

Check if the same lab product or an alternative is used in the 5 most similar protocols
Blood was collected from the eyes of the mice and left to stand for 30 min, after that it was centrifuged at 4°C at 1000 g for 10 min. After the serum was collected, the TP and ALB concentrations were analyzed using an automatic biochemical analyzer (BS‐350E, Shenzhen Mindray Bio‐medical Electronics Co., Ltd.).

Measuring Serum Lipid Levels

Check if the same lab product or an alternative is used in the 5 most similar protocols
Serum levels of TG, TC, and LDL-C were measured by the certified core clinical laboratory at the No. 3 Hospital of Beijing University, who used the respective commercial kits and Chemistry Analyzer (BS-350E) provided by Shenzhen Mindray Biomedical Electronics Co., Ltd. (Shenzhen, China) [75 (link)].

Routine Blood Analysis in Mice

Check if the same lab product or an alternative is used in the 5 most similar protocols
For blood routine examination, 50 μL fresh blood was collected from each mouse and mixed with EDTA immediately. The blood samples were analyzed by Celltac Alpha MEK-6400 series hematology analyzers (Nihon Kohden, MEK-6400). For serum biochemical analysis, blood samples were collected, clotted for 2 h at room temperature or overnight at 4 °C, and then centrifuged (1000× g, 10 min) to obtain serum. 200 μL serum was aliquot and analyzed of Alanine transaminase (ALT), aspartate transaminase (AST), uric acid (UA) and creatinine (CREA) by Chemistry Analyzer (Mindray, BS-350E).

Vaccine Safety Evaluation in Mice

Check if the same lab product or an alternative is used in the 5 most similar protocols
For vaccine safety evaluation, C‐CPS was subcutaneously injected into healthy BALB/c mice with 20 μg of polysaccharide. After 1 h, the mouse body weight and rectal temperature were measured at different time points, and the sera were collected at the same time. Cytokines (IL‐1β, TNF‐α, IL‐6 and IFN‐γ) were detected using commercially available ELISA kits (Dakewe, Shenzhen, China) according to the manufacturer’s instructions. The serum levels of alanine aminotransferase, aspartate transaminase, alkaline phosphatase, lactate dehydrogenase and blood urea nitrogen were detected using a biochemical autoanalyser (BS‐350E, Mindray).

Hematological Examination of Drug Toxicity

Check if the same lab product or an alternative is used in the 5 most similar protocols
Hematological examination was performed to explore the potential toxicity of drugs in vivo. BALB/c nude mice were divided into four groups (n = 5), and injected by NS, naked siRNA (0.3 mg/kg), GCE/siRNA (containing 0.3 mg/kg siRNA), and DOX (1.2 mg/kg) every other day for 5 times. After the drug administration, blood samples were collected from all nude mice, and blood routine test was performed by an automatic hematology analyzer (MEK6400, Nihon Kohden, Japan).
The whole blood was taken from the blood collection vessel without anticoagulant for blood biochemical examination. The whole blood was kept at room temperature for 2 h, and centrifuged (3 000 × g, 4 °C) for 10 min to obtain the upper serum. The important blood biochemical indicators of liver function (ALT: alanine aminotransferase, AST: aspartate aminotransferase, ALP: alkaline phosphatase, TB: total protein, and ALB: albumin) and renal function (CREA: creatinine, UA: uric acid, and UREA) of nude mice were detected by an automatic biochemical analyzer (BS-350E, Mindray, China).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
