Immunoblotting Analysis of Immune Proteins

Immunofluorescent Analysis of Proteasome Inhibition

Antibodies to β2i (ab77735), β2 (ab22650), and α-smooth muscle actin (α-SMA) (ab5694) were purchased from Abcam Inc. Antibodies to Mac-2 (sc-20157), myocardin (sc-33766), GAPDH (sc-365062) and 4′,6-diamidino-2-phenylindole (DAPI) were from Santa Cruz Biotechnology. Antibodies to CD4 (NBP1-19371) were from Novus. All antibodies were used in a 1:200 dilution. For immunofluorescent staining, antibodies to β5i (ab3329), pCNA (ab18197) and α-SMA were from Abcam Inc. Anti-mouse or anti-rabbit conjugated antibodies were from Cell Signaling Technology. TUNEL assay kit was from Promega. Proteasome Subunit Quantification by Western Blot

Proteins were transferred to PVDF membranes using a Bio-Rad mini-protean transfer system containing a buffer of 25 mM Tris base, 192 mM glycine, 0.1% SDS. Proteins were transferred for 6h at 40V at 4°C. Membranes with transferred proteins were blocked with a 1:1 mixture of Li-COR blocking buffer and PBS. Membranes were then incubated in blocking solution for 1 h and primary Abs (1:1000) applied for 16 h at 4°C. Membranes were washed and incubated with secondary Abs (IR Dye® 800CW goat anti-mouse IgG (H+L) or IR Dye 680CW donkey anti-rabbit IgG (H+L), and imaged using an Odyssey Li-Cor platform.
