Materials were purchased from standard sources: luteolin from Sigma (St Louis, MO, USA); MTT from Sigma; DMEM and FBS from Gibco BRL (Waltham, MA, USA); methanol and acetic acid (HPLC grade) from Fisher Scientific  (Waltham, MA, USA); and dimethyl sulfoxide (DMSO) and acetone from KeLong Chemicals (Chengdu, China). Antibodies used include rat anti-mouse CD31 polyclonal antibody (BD Pharmingen™, Franklin Lake, NJ, USA), rabbit anti-mouse PCNA antibody (Abcam, Cambridge, UK), and rhodamine-conjugated secondary antibody (Abcam). NH2-PEG-PCL was purchased from ruixi Biological Technology Company (Xian, China).
C57 mice (6–8 weeks) and Sprague Dawley (SD) rats were purchased from the Animal Center Laboratory of Beijing HFK Bioscience Co Ltd (Beijing, People’s Republic of China). All mice were housed in a controlled temperature of 20–22°C, with a relative humidity of 50–60% and 12 hr light-dark cycles. The animals were provided with standard laboratory chow and tap water ad libitum. All animal procedures were performed following the protocol approved by the Institutional Animal Care and Treatment Committee of Sichuan University (Chengdu, People’s Republic of China) and treated humanely throughout the experimental period.

Hypoxic Tumor Microenvironment Cell Lines

Murine Lewis lung cancer cell (LLC) and human NSCLC cell H1975 (EGFR T790M, L858R) were purchased from American Type Culture Collection (ATCC). Mouse microglia BV2 was purchased from Procell Life Science & Technology CO.,Ltd (Wuhan, China). HCC827 (human lung adenocarcinoma cell line, EGFR exon 19 deletion) was acquired from the Institute of Biochemistry and Cell Biology of the Chinese Academy of Sciences(Shanghai, China). Beas-2B (human lung epithelial cell line) and PC-9 (human lung adenocarcinoma cell line, EGFR exon 19 deletion) were maintained in the laboratory of cancer center of Union Hospital, Tongji Medical College, Huazhong University of Science and Technology. BV2 and LLC cells were cultured in Dulbecco's Modified Eagle Medium (DMEM) supplemented with 10% fetal bovine serum (FBS) at 37 °C in a humidified incubator with 5% CO2. DMEM and FBS were purchased from Gibco (Carlsbad, USA). HCC827, H1975, Beas-2B and PC-9 cells were cultured in RPMI 1640 medium (Gibco) supplemented with 10% FBS. Cells were grown at 37 °C in a humidified atmosphere of 5% CO2, and cells at the logarithmic phase were used in the experiments. To mimic hypoxia tumor environment, cells were incubated in a hypoxic incubator to create a hypoxic microenvironment. KC7F2 (MCE, 40 μM) was used as a HIF-1 inhibitor. The exact concentration details have been indicated in the corresponding figure.

Huh7 Cell Culture Protocol

Human HCC Huh7 cells were purchased from the Cell Bank, Chinese Academy of Sciences (Shanghai, China). The normal (non-cancerous) human liver cells (hepatocytes) with normal functionalities were used as a control for all the experiments. Dulbecco’s modified Eagle’s medium (DMEM) supplemented with 10% FBS (DMEM and FBS from Gibco, Shanghai) and antibiotics were used for cell culturing at 37°C and 5% CO2.
