AOM-DSS Colorectal Cancer and Colitis Models

For the CA-CRC model19 (link), 8–10 week-old C57BL/6J, A/J, or Irf1−/− mice (The Jackson Laboratory) were injected ip with azoxymethane (AOM; 7 mg/kg) followed by three 4-day cycles of 2% (w/v) DSS in the drinking water, with each cycle 17 days apart and euthanized 6 weeks later. In some experiments, mice were injected with AOM (7 mg/kg) once, followed by two 4-days cycles of 2% (w/v) DSS and sacrificed 3 weeks later. The model of AOM-induced colorectal cancer was as previously described20 (link). For the chronic colitis model, mice were treated with three 4-days cycles of 2% (w/v) DSS and sacrificed after week 8.

Harvesting Organs from Genetically-Modified Mice

C57BL/6J (BL6) and IRF-1−/− mice (Matsuyama et al., 1993 (link)) were obtained from The Jackson Laboratory (Bar Harbor, Maine). IFNAR1−/− mice on the BL6 background were a gift from Dr. Mitchell Grayson (Grayson et al., 2007 (link)). IRF-3−/− and IRF-7−/− mice were a gift from Dr. Michael Diamond (Honda et al., 2005 (link); Sato et al., 2000 (link)). All mice utilized in this study are on the C57BL/6J background. Organs were harvested from naïve mice between 8 and 12 weeks of age. Mice were housed and bred in a specific-pathogen-free barrier facility in accordance with federal and institutional guidelines. All experimental manipulations of mice were approved by the Institutional Animal Care and Use Committee of the Medical College of Wisconsin.

CKD Model and IS Treatment in Mice

IRF1 knockout (IRF1-/-) mice were purchased from Jackson Laboratory (Bar Harbor, ME, USA). To construct a CKD model, 8-week-old male Balb/c mice received electrocoagulation of right 2/3 renal cortex first and left total nephrectomy two weeks later, as previously described 16 (link). CKD mice were fed a diet containing 5% AST-120 (Kremezin, Kure ha Chemical Industry, Tokyo, Japan) for 8 weeks. WT C57BL/6J and IRF1-/- mice were intraperitoneally injected with IS (100 mg/kg, Sigma-Aldrich, St. Louis, MO, USA) daily for 8 weeks. Each group included at least 8 mice. All animal procedures were approved by the Committee of Ethics on Animal Experiments of the Army Medical University.
