ELISA for Dengue and Zika Antibodies

Check if the same lab product or an alternative is used in the 5 most similar protocols
Capture ELISA assay was performed by coating a 96 well plate with DENV-4, DENV-2 (Fitzgerald), or ZIKV (MyBiosource) antigen 2.5 ug/mL coating buffer (Sigma, 08058) overnight at 4°C. Unbound antigen was washed with PBS containing 0.05% Tween 20 and further blocked with 5% BSA (Fisher). Serum samples were serially diluted (1:100, 1:3) in blocking buffer and incubated for 1 h at 37°C. Unbound antibodies were removed by washing and incubated for 1 h at 37  °C with goat-anti-human secondary Ab conjugated with horseradish peroxidase (HRP) (Bio-Rad, CA). Unbound secondary Ab was washed off, and signals were developed with o-phenylenediamine dihydrochloride substrate tablets (Sigma, 34006). OD was read at 492 nm.

Virus Capture ELISA Assay Protocol

Check if the same lab product or an alternative is used in the 5 most similar protocols
For virus capture ELISA assay, Microlon 600 96-well plates (VWR, Radnor, PA) were coated overnight at 4 °C with a mixture of 100 ng of 4G2/2H2 Abs in 0.1 ml coating buffer (Sigma, 08058). Unbound Abs were washed off with PBS containing 0.05% Tween 20, and blocked with 3% BSA (Fisher). Virus was diluted in blocking buffer to optimal concentrations, and 0.1 ml was added to each well and incubated 1 h at 37 °C. Unbound viruses were removed by washing, and serum samples diluted in blocking buffer were added and incubated for another hour. Unbound serum was removed by washing and then incubated for 1 h at 37 °C with goat-anti-monkey secondary Ab conjugated with HRP (Fitzgerald, MA, USA). Unbound secondary Ab was washed off, and signals were developed with o-phenylenediamine dihydrochloride substrate tablets (Sigma, 34006). OD was read at 492 nm.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
