Microbiological Analysis of Dairy Samples

Dairy samples -milk, whey, buttermilk, cream, and dairy wastewater (raw effluent P1, raw effluent averager, raw effluent -centrifuge shot and treated effluent after flotation) were analyzed. Before microbiological cultures were performed, appropriate dilutions of 10 -1 , 10 -2 , 10 -3 were made for samples of milk, whey, buttermilk, cream, and for dairy wastewater 10 -3 and 10 -4 , respectively. In the next step, surface cultures were performed on 5 selected culture media: All Purpose Tween Agar (APT Agar), M-17 Agar, de MAN, ROGOSA and SHARPE Agar (MRSA) with 1% Tween80 (Sigma Aldrich, Germany), China Blue Lactose Agar (CBL), Milk Plate Count Agar (MPCA). Therefore, 100 µl of each sample was taken for this purpose and then inoculated on Petri dishes (Alchem, Poland). Most of the culture media came from Sigma Aldrich (Germany), and only the MPCA from Graso Biotech (Poland). The media were incubated for 24 h under aerobic conditions at 37°C (APT Agar, M-17 Agar, China Blue Lactose, MPCA) and under anaerobic conditions at 37°C (MRSA). To obtain pure bacterial cultures, a serial dilution method, solid medium culture and, as a last resort, reduction culture was performed.

Other chemicals used in the study were ammonium chloride (VWR), potassium phosphate dibasic anhydrous, sodium phosphate monobasic monohydrate, ferrous chloride tetrahydrate, sodium selenite, sodium molybdate dihydrate, nickel chloride hexahydrate, zinc chloride, manganese chloride tetrahydrate, cobalt (II) chloride hexahydrate, copper (II) chloride dihydrate, boric acid, hydrochloric acid, EDTA disodium salt dihydrate, sodium hydroxide, lactate, acetate, l-cysteine HCl (Sigma), Bacto agar, resazurin (Difco), milk plate count agar and maximum recovery diluent (Sigma).
