Quercetin-Based Compound Characterization

The compound Quercetin (QUA) [2-(3,4-dihydroxyphenyl)-3,5,7-trihydroxychromen-4-one], with a purity of 98%, extra pure grade yellow color, and a CAS number of 6151-25-3, was acquired from Sisco Research Laboratories Pvt. Ltd. (Mumbai, India). Copper sulfate pentahydrate (CuSO4·5H2O) and Tween 80 were obtained from Merck (Mumbai, India). The reference/standard drugs Ondansetron (OND), Hyoscine butyl bromide (HYS), and Domperidone (DOM) were obtained from Incepta Pharma. Ltd. (Dhaka, Bangladesh), Opsonin Pharma. Ltd. (Barishal, Bangladesh), and Beximco Pharma. Ltd. (Dhaka, Bangladesh), respectively, all located in Bangladesh.

Quantifying Flavonoid Content in Extracts

The total flavonoid content was determined using the Dowd method as adapted by Arvouet-Grand [26 (link)] and reported by others [16 (link),27 ,28 (link)]. Briefly, 2 mL of 2% aluminium trichloride (AlCl3) (Changzhou Baixing, Tianjin, China) in methanol was mixed with equal volume of the extract and incubated at room temperature for 10 min before reading the absorbance at 415 nm against a blank sample consisting of 2 mL of extract and 2 mL of methanol without AlCl3. The total flavonoid content was determined using a standard curve with quercetin (Sisco Research Laboratories, Mumbai, India) as the standard. The mean of three readings was expressed as milligrams of quercetin equivalent per gram of the weight of extract (mg QE/gDW).
