Formaldehyde 37% (Vetec, Brazil), morphine hydrochloride (Vetec, Brazil), caffeine (Merck, USA), and naloxone hydrochloride (Cristália, Brazil) were also purchased and used in this study. All drugs were diluted in distilled water except 2-AP which required Tween 80 and distilled water. The other solvents and chemicals used in this study were purchased from Sigma-Aldrich® (São Paulo, São Paulo, Brazil).

Pharmacological Agents in Research

The following drugs and reagents were acquired in Sigma-Aldrich® (St. Louis, MO, United States): 5-fluorouracil (5-FU), diazepam, naloxonazine, cyclophosphamide, and Tween 80. The anesthetics ketamine and xylazine were obtained from Konig S.A. (Brazil), and MSD Saúde Animal (Brazil), respectively. Other drugs, reagents, and kits comprise: heparin (HEPAMAXS®), ELISA Kit (eBioscience®, Inc. Science Center Drive, San Diego, CA, United States), kits for hematological and biochemical analysis (LABTEST; Lagoa Santa, MG, Brazil), morphine hydrochloride (Merck®), naloxone hydrochloride (Cristália®), formaldehyde 37% (Vetec), dimethylsulfoxide - DMSO (Mallinckrodt Chemicals - Phillipsburg, NJ, United States). The acridine derivative N’-(6-chloro-2-methoxy-acridin-9-yl)-2-cyanoacetohydrazide (ACS-AZ) was synthesized as previously described (Silva et al., 2018 (link)).

Intraperitoneal Drug Administration Protocol

Genotoxicity Assay of DFMC in Sarcoma Cells

Sarcoma 180 cells were plated in 24-well plates (0.5 × 106 cells/mL) and treated with DFMC at 5, 25, and 50 μg/mL. After 44 h at 37°C, cytochalasin B (Sigma Aldrich, USA, 6 μg/mL) was added, and the cells were reincubated for an additional 28 h. At 72 h, the cultures were transferred to tubes and centrifuged at 800 rpm for 5 minutes. Then, the supernatant was removed, and the body of the cell bottom was enlarged and centrifuged again before the addition of 2 mL of fixing solution (methanol and acetic acid, ratio 5 : 1) and 3 drops of formaldehyde 37% (Vetec™, Brazil). This procedure was repeated 3x using fixing solution 3 : 1 without formaldehyde. Supernatants were discarded, and 2-3 drops of cell suspension were dripped onto slides and stained with Giemsa for 5 min [25 (link)]. Considering blind examination, a total of 2000 cells by concentration were counted by optical microscopy at 1000x (1000 cells/slide) to count buds, bridges, and micronuclei.
