Purification and Characterization of Sulfotransferases

7-dehydrocholesterol (7-DH), 24-dehydrocholesterol (24-DH), dithiothreitol (DTT), ethylene-diamine-tetraacetic acid (EDTA), L-glutathione (reduced), 1-hydroxypyrene (1-HP), imidazole, isopropyl-thio-β-D-galacto-pyranoside (IPTG), lysozyme, 3-maleimido-PROXYL, N-cyclohexylmaleimide, pepstatin A, potassium phosphate, 2,2,2-trichloroethanol (TCE), and quercetin were the highest grades available from Sigma. Ampicillin, dimethyl sulfoxide (DMSO), KOH, LB media, MgCl2, tris(hydroxymethyl)aminomethane (Tris) base, and phenylmethylsulfonyl fluoride (PMSF) were purchased from Fisher Scientific. Glutathione- and nickel-chelating resins and Superdex 200 Increase, 10/300 GL columns were obtained from GE Healthcare. D2O was purchased from Cambridge Isotope Laboratories. Competent E. coli (BL21(DE3)) was purchased from Novagen. SULT2B1a and SULT2B1b clones were obtained from IDT. The SULT2A1 clone and SULT2B1b pKK233 (32 (link)) vector were obtained from Dr. C. N. Falany. Synthesis and purification of PAPS and PAP is previously described (33 (link), 34 (link)) and anion-exchange HPLC analysis suggested they are ~99% pure.

are as follows. Adenosine
monophosphate (AMP), dithiothreitol (DTT), dimethyl sulfoxide (DMSO),
ethylenediaminetetraacetic acid (EDTA), 1-hydroxypyrene (1-HP), 4-hydroxytamoxifen
(afimoxifene, TAM), imidazole, isopropyl thio-β-d-galactopyranoside
(IPTG), Luria broth (LB), lysozyme, β-mercaptoethanol (β-ME),
pepstatin A, Na2HPO4, and NaH2PO4 were obtained from Sigma. Ampicillin, HEPES, KCl, KOH, MgCl2, NaCl, and phenylmethanesulfonyl fluoride (PMSF) were purchased
from Fisher Scientific. Glutathione- and nickel-chelating resins were
obtained from GE Healthcare. Competent Escherichia coli [BL21(DE3)] cells were purchased from Agilent Technologies. PAP,
PAPS, and [35S]PAPS were synthesized in house as previously
described12 (link),16 (link),17 (link) and were ≥98%
pure as assessed by anion-exchange high-performance liquid chromatography.

Sulfotransferase Enzyme Characterization

Acetaminophen (Acet), apomorphine (AP), dithiothreitol (DTT), dehydroepiandrosterone (DHEA), ethylenediaminetetraacetic acid (EDTA), 17-β-estradiol (E2), fulvestrant (Ful), L-glutathione (reduced), 1-hydroxypyrene (1-HP), imidazole, isopropyl thio-β-D-galactopyranoside (IPTG), Lysogeny broth (LB), lysozyme, β-mercaptoethanol, 1-naphthol (1-Nap), 2-naphthol (2-Nap), pepstatin A, resveratrol (Res), and sodium phosphate were the highest grade available from Sigma. PAPS and [35S]PAPS were synthesized in house21 (link) and were >98% pure. Ampicillin, HEPES, KOH, MgCl2, NaCl, KCl, LiCl, and phenylmethanesulfonyl fluoride (PMSF) were purchased from Fisher Scientific. Glutathione- and nickel-chelating resins and PreScission protease were obtained from GE Healthcare. Competent Escherichia coli [BL21(DE3)] was purchased from Novagen. The QuikChange mutagenesis kit was purchased from Agilent Technologies, and mutagenic primers were purchased from Fisher Custom Oligos. An Amicon Ultra Centrifugal Filter [molecular weight cutoff (MWC) of 10000] was purchased from EMD Millipore. PEI-F anion exchange TLC sheets were purchased from Merck, KGaA.

Ampicillin, HEPES, KCl, KOH, MgCl2, NaCl and phenylmethylsulfonyl fluoride (PMSF) were purchased from Fisher Scientific. Epigallocatechin gallate (EGCG) and epigallocatechin (EGC) were obtained from Santa Cruz Biotechnology, Inc. Anion exchange HPLC was performed using an Eprogen, AX100 (5μm) column. Glutathione- and nickel-chelating resins were obtained from GE Healthcare. Competent E. coli (BL21(DE3)) was purchased from Novagen. PAPS and PAP were synthesized in house as previously described (16 (link), 31 (link)) and were > 98% pure as assessed by anion-exchange high- performance liquid chromatography.

Dehydroepiandrosterone (DHEA), the chemical, was purchased from TCI America, DHEA [1,2,6,7-3H(N)] was obtained from PerkinElmer, DHEA, the nutraceutical, was obtained from the General Nutrition Corporation, lot 57429B17. Mefenamic acid, the chemical, was obtained from Santa Cruz Biotechnology. Mefenamic acid, the drug, was obtained from Lupin Limited, Lot G700974. Deuterium Oxide (> 99%) was purchased from Acros Organics. Glutathione- and nickel-chelating resins were obtained from GE Healthcare. Competent E. coli (BL21(DE3)) was purchased from Novagen. PAPS was synthesized as previously described [17 (link),18 (link)] and was ≥99% pure as assessed by anion-exchange HPLC.
