Antibiotic Susceptibility Testing Protocol

Exebacase MICs were determined by broth microdilution using CAMHB-HSD medium (16 (link), 18 , 20 (link)), comprised of BBL Mueller-Hinton II (cation-adjusted) broth (catalog no. 212322, lot 1242967; Becton, Dickinson, Franklin Lakes, NJ) with horse serum (25% [vol/vol]) (catalog no. H1270, lot no. 21C414; Sigma-Aldrich, St. Louis, MO) and 0.5 mM dl-dithiothreitol (pH 7.2 to 7.4) (Sigma-Aldrich). As described in CLSI standard M07-A11 (20 (link)), daptomycin MICs were determined in CAMHB supplemented to 50 μg/mL with CaCl2, oxacillin MICs were determined in CAMHB with 2% NaCl, and vancomycin MICs were determined in CAMHB. In addition, oxacillin, daptomycin, and vancomycin MICs were generated against each strain using CAMHB-HSD. MIC panels were incubated for 18 to 20 h at 37°C.
