Culturing Human Melanoma Cell Lines

The human melanoma A375-S2, SKMEL-28, SKMEL-5, MeWO and RPMI-7951 cell lines were purchased from the National Rodent Laboratory Animal Resource (Shanghai, China). All melanoma cell lines were grown in Gibco Dulbecco's modified Eagle's medium (Thermo Fisher Scientific, Inc., Waltham, MA, USA) supplemented with 10% fetal bovine serum (FBS; Sigma-Aldrich, St. Louis, MO, USA) and 100 units/ml Invitrogen penicillin-streptomycin (Thermo Fisher Scientific, Inc.). Normal human epidermal melanocytes (NHEMs) from adult skin (PromoCell GmbH, Heidelberg, Germany) were maintained in serum- and phorbol myristate acetate-free melanocyte growth medium M2 (PromoCell GmbH). The cell lines were cultured in a humidified incubator at 37°C in a 5% CO2 and 95% air atmosphere for 2–3 days.

MRC-5 Fibroblast Cell Culture and miRNA Transfection

The MRC‐5 human lung fibroblast cell lines were purchased from the Chinese Academy of Sciences Shanghai Branch (Shanghai, China). MRC‐5 cells were grown in Dulbecco’s Modified Eagle’s Medium (DMEM; Thermo Fisher Scientific, Inc., Waltham, MA, USA) supplemented with 10% fetal bovine serum (FBS; Sigma‐Aldrich, St. Louis, MO, USA) and 100 units/ml Invitrogen penicillin–streptomycin (Thermo Fisher Scientific, Inc.). The cells were cultured in a humidified incubator at 37˚C in 5% CO2 and 95% air atmospheres for 2–3 days. miRNA transfection was performed using Lipofectamine 2000 in accordance with the manufacturer's guidelines (Invitrogen). Untreated cells were designated as the control group.
