Quantification of Buprenorphine in Rabbit Plasma

Analytical grade buprenorphine was purchased from LGC Standards (Teddington, UK) and buprenorphine-D4 was purchased from Sigma-Aldrich (Darmstadt, Germany). Ethyl acetate (POCH, Gliwice, Poland), n-hexane (Merk, Darmstadt, Germany) and ammonia solution 25% (Stanlab, Lublin, Poland) were of HPLC or analytical grade. Formic acid and methanol were from Sigma-Aldrich (Darmstadt, Germany) and were of analytical grade and LC-MS grade, respectively.
The plasma samples used in the method development and validation were obtained from rabbits involved in the pharmacokinetic/pharmacodynamic study. All blank plasma samples were obtained from animals which did not receive any drugs except isoflurane used in inhalatory anesthesia.

Opioid Receptor Ligand Characterization

U-47700 and furanyl fentanyl were procured from Cayman Chemical. Hydromorphone hydrochloride was obtained from Fagron. Buprenorphine, norBuprenorphine, fentanyl, loperamide, carfentanil, and naloxone were obtained from LGC Standards. Dulbecco's modified Eagle's medium, Opti-MEM I Reduced Serum Medium, penicillin and streptomycin (5000 IU/mL and 5000 g/mL), amphotericin B (250 g/mL), glutamine (200 mmol/L), the restriction enzyme SacI, and the DNA polymerase (Phusion polymerase) were purchased from Thermo Fisher Scientific. Primers were procured from Eurofins Genomics. [d-Ala 2 -MePhe 4 -Gly-ol]encephalin (DAMGO), fetal bovine serum (FBS), and poly-D-lysine were from Sigma Aldrich. Analytical Standards for Pharmaceutical Compounds

Extraction and separation solvents, methanol (MeOH) and acetonitrile (AcN) were purchased from Fisher-Scientific, assuming an analytical grade (purity up to 99.95 %).
