Caspase-3 activity was determined using Alexa Fluor 647-conjugated anti-active caspase-3 antibody (BD Biosciences, San Diego, CA, USA) according to the manufacturer’s instructions. Briefly, HCT116 and H460 cells (1 × 106) were trypsinized, pelleted, washed twice with ice-cold PBS and fixed in Fixation Buffer for 20 min on ice. After fixation, cells were rinsed twice with Perm Wash Buffer, stained with Alexa Fluor 647-conjugated anti-active caspase-3 antibody for 30 min, rinsed with Perm Wash Buffer and analyzed by flow cytometry with FACS Accuri C6 (BD, San Jose, CA, USA).
