Investigating the Effects of Synthetic Cannabinoids

Δ9-THC, JWH-018, JWH-073, JWH-081, JWH-210, rimonabant, and SR144528 were obtained from the National Institute on Drug Abuse (NIDA, Bethesda, MD) through the NIDA Drug Supply Program. JWH-167 and JWH-391 were synthesized by John Huffman (Clemson University, Clemson, SC). AM-2201 was purchased from Cayman Chemical (Ann Arbor, MI). For in vivo administration, all cannabinoid compounds were dissolved in a vehicle of 7.8% Polysorbate 80 N.F. (VWR, Marietta, GA) and 92.2% sterile saline USP (Butler Schein, Dublin, OH). Mianserin HCl was obtained from Sigma Aldrich (St. Louis, MO) and was dissolved in sterile saline USP. All compounds used in the FOB procedure were administered intravenously (i.v.) into the tail vein at a volume of 10 ml/kg. Details of compound administration for the drug discrimination procedure are presented in section 2.6.

Characterization of Synthetic Cannabinoids

All solvents were high-performance liquid chromatography (HPLC) grade, purchased from Fisher Scientific (Fair Lawn, NJ), EMD Chemical (Gibbstown, NJ), and Sigma-Aldrich (St. Louis, MO). JWH-018, AM2201, JWH-122, UR144, XLR11, and 5F-AKB48 reference standards were obtained from Cayman Chemical (Ann Arbor, MI). JWH-019 reference standard was synthesized in the laboratory of Dr. John W. Huffman and provided by Dr. Jenny L. Wiley. Herbal products were obtained from the Richland County Sheriff’s Department (Columbia, SC). These products were seized between December 2011 and August 2013 and were scheduled for destruction. Custody was turned over to RTI International, and products were stored in accordance with established protocols.

Characterization of AM-2201 Metabolism

AM-2201 was purchased from Cayman Chemical Company (Ann Arbor, MI, USA). Acetaminophen, alamethicin, chenodeoxycholic acid, coumarin, diclofenac, 7-hydroxycoumarin, midazolam, mycophenolic acid, N-acetylserotonin, naloxone, naloxone 3-β-d-glucuronide, NADPH, phenacetin, trifluoperazine, Trizma® base, and uridine 5′-diphospho-glucuronic acid (UDPGA) were purchased from Sigma-Aldrich (St. Louis, MO, USA). Ultrapool human liver microsomes (150 donors), 13C2,15N-Acetaminophen, bufuralol, N-desethylamodiaquine, 1′-hydroxybufuralol, 4-hydroxy-diclofenac, 4-hydroxymephenytoin, d3-4-hydroxymephenytoin, 1′-hydroxymidazolam, d9-1-hydroxybufuralol, and [S]-mephenytoin were obtained from Corning Life Sciences (Woburn, MA, USA). SN-38 was obtained from Santa Cruz Biotechnology (Dallas, TX, USA). N-Acetylserotonin β-d-glucuronide, chenodeoxycholic acid-24-acyl-β-glucuronide, mycophenolic acid β-d-glucuronide, and SN-38 glucuronide were obtained from Toronto Research Chemicals (Toronto, ON, Canada). Acetonitrile and methanol (high performance liquid chromatography (HPLC) grade) were obtained from Fisher Scientific (Fair Lawn, NJ, USA). All other chemicals were of the highest quality available.

Analytical Characterization of Synthetic Cannabinoids

All reagents were of analytical reagent grade unless otherwise stated. Ethanol (EtOH), formic acid, p-dimethylamino benzaldehyde (p-DMAB), sulfuric acid, salicylic acid and urea were purchased from Sigma-Aldrich (St. Louis, MO, USA). Mannitol and silica gel 60 (0.063 -0.200 mm), as well as acetonitrile and methanol for chromatography were obtained from Merck (Darmstadt, Germany). Certified reference materials; AM-2201
were purchased from Cayman Chemicals (Ann Arbor, MI).
The non-certified synthetic cannabimimetics; XLR-11 [1-(5-fluoropentyl)-1H-indol-3-yl](2,2,3,3-tetramethylcyclopropyl)methanone and 5F-PB-22 (quinolin-8-yl 1-(5-fluoropentyl)-1Hindole-3-carboxylate) were obtained from Istanbul Police Directorate Crime Laboratory.
