Protease inhibitor and phosphatase inhibitor 1 and 2

Manufactured by Merck Group
Sourced in United States

Protease inhibitor and phosphatase inhibitor I and II are laboratory reagents used in various research applications. Protease inhibitor is a chemical compound that blocks the activity of proteases, while phosphatase inhibitor I and II are used to inhibit the activity of phosphatases. These reagents are commonly used in protein purification, cell biology, and biochemical assays to preserve the integrity of target proteins or enzymes.

Immunoprecipitation of CoREST from Hippocampal Neurons

Check if the same lab product or an alternative is used in the 5 most similar protocols
Hippocampal neuron lysates were generated by using lysis buffer (Cell Signaling, MA, USA) contained protease inhibitor and phosphatase inhibitor I and II (Sigma). 2 μg antibody (CoREST, Millipore, CA, USA, #07-455) or rabbit IgG (Millipore, CA, USA) was added to lysates containing 500 μg protein at 4°C overnight [12 (link)], which was followed by immunoprecipitation with 30 μl Protein G Agarose beads (Roche, Mannheim, Germany). Beads were washed five times with HNTG buffer (20 mM HEPES pH 7.5, 150 mM NaCl, 0.1% Triton X-100 and 10% glycerol). The bound proteins were eluted with 2X Laemmli sample buffer (126 mM Tris-HCl, 20% glycerol, 4% SDS, 0.02% bromophenol blue) and analyzed by Western blotting.

Inhibition of Neuronal Activity

Check if the same lab product or an alternative is used in the 5 most similar protocols
The following inhibitors were used: protease inhibitor and phosphatase inhibitor I and II (Sigma), volasertib (BI6727) (Selleckchem). Picrotoxin (PTX) (Acros) was freshly prepared as a 10 mM stock in 0.1 M NaOH and treated at 25 μM final concentration in hippocampal cultured neurons for 18–20 hours.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
