Quantification of Hypothalamic β-endorphin

Ex vivo measurements of β-endorphin content in mediobasal hypothalamus derived from male WT and corresponding Cnr1−/− littermates was assessed by use of a commercially available β-endorphin ELISA Kit and in accordance to manufacturer’s instructions (MyBioSource, MBS703919; San Diego, CA, USA). Briefly, 30 mg of tissue was rinsed and mechanically homogenized in PBS. After two freeze-thaw cycles, the homogenates were centrifuged for 5 min at 5000 × g and 4° C. The supernatant was removed and assayed immediately. In parallel, sample protein concentrations were measured using the BCA kit (Thermo scientific, Cat# 23228 and 1859078), and β-endorphin content was calculated as ng β-endorphin / mg protein.

Quantifying β-endorphin Levels in hSOD1G93A Mice

The hypothalamus of hSOD1G93A mice treated with vehicle or TRAM‐34 + rimonabant was disrupted with a homogenizer and analysed for β‐endorphin content using a commercially available β‐endorphin elisa kit, in accordance to manufacturer's instructions (MyBioSource). Briefly, after two freeze–thaw cycles were performed to break the cell membranes, the homogenates were centrifuged for 5 min at 5000× g, 2–8°C. The supernatant was removed and assayed immediately. BCA Reagent Kit was used to measure protein concentration in samples (Thermo Scientific) and β‐endorphin content was calculated as pg·ml−1.
