SARS-CoV-2 Inhibition Assay using Cellular Enzymes

The compound antipain (i.e., omicsynin B4) was purchased from the Efebio Inc. (Shanghai, China) and the purity was above 97% based on HPLC. The inhibitors in this study were used as follows. Ribavirin (RBV) was purchased from Sigma-Aldrich (St. Louis, MO, USA). Camostat mesylate, nafamostat mesylate, E−64d, remdesivir (RDV), ammonium chloride (NH4Cl), hexa-D-arginine, bivalirudin TFA, argatroban monohydrate, ebselen and boceprevir were purchased from MedChemExpress (Monmouth Junction, NJ, USA). All of compounds were greater than 95% purity and prepared in DMSO, and the concentrations were described in the figure legends.
Human CTSL expression plasmid (pCMV3-CTSL) and empty control vector (pCMV3) were purchased from Sino Biological Inc. (Beijing, China). pcDNA3.1(+)-TMPRSS2, pcDNA3.1(+)-Furin and pcDNA3.1(+) empty control vector were synthesized and provided by Genscript Inc. (Nanjing, China).
