Quantification of Ligandrol (LGD-4033) and Metabolites

The standard of ligandrol (LGD-4033) was purchased from Toronto Research Chemicals; TRC (Toronto, ON, Canada). Stock solution of standard substance was prepared at the concentration of 1 mg/mL and its working solutions (100 µg/mL, 10 µg/mL, and 1 µg/mL) in methanol and stored at −20 °C.
Due to unavailability of certified reference materials of LGD-4033 metabolites, the reference urine collected from volunteers who ingested 50 µg of LGD-4033 dissolved in 120 mL of drinkable yogurt was utilized in the experiment [14 (link)].
LC/MS-grade of methanol and acetonitrile were purchased from MerckMillipore (Darmstadt, Germany) and Fisher Chemical (Hampton, NH, USA), respectively. Disodium phosphate and monosodium phosphate ≥ 99% purity were supplied by Honeywell (Charlotte, NC, USA), while potassium carbonate and potassium bicarbonate, both HPLC grade, were purchased at POCH S.A. (Gliwice, Poland). In turn, β-glucuronidase (Escherichia coli) and HPLC-grade methyl tert-butyl ether were supplied by Roche (Basel, Switzerland) and J.T.Baker (Philipsburg, NJ, USA), respectively. The Millipore DirectQ UV3 system (Darmstadt, Germany) was used as the source of water (R > 18 MΩ/cm). Sample preparation of dietary supplement samples followed the protocol described in [19 (link),20 (link)].

Quantitative Analysis of Ginsenosides

Quantification of NVP and Metabolites

Standard, NVP was attained from TargetMol (Shanghai, China). Furthermore, 2-OH NVP and 3-OH NVP were supplied by Toronto Research Chemicals (Toronto, ON, Canada). NVP-d3 as internal standard (IS) of NVP and 2-OH NVP-d3 as IS of 2-OH NVP and 3-OH NVP were supplied by Toronto Research Chemicals (Toronto, ON, Canada). Ammonium acetate of LC-MS grade was obtained from Tedia (Fairfield, OH, USA), methanol of LC-MS grade was obtained from Sigma Aldrich (St. Louis, MO, USA). Deionized water was purchased from A.S. Watson Group (Hong Kong, China).
NVP, 2-OH NVP and 3-OH NVP were separately dissolved in methanol as stock solutions at concentrations of 1 mg/mL, 0.5 mg/mL and 50 μg/mL, respectively. The mixed working solution of the three compounds was needed to dilute the stock solution to the expected concentrations with methanol, such as 0.8–10,000 ng/mL for NVP and 0.4–250 ng/mL for 2-OH NVP and 3-OH NVP. The IS working solutions with high concentration were diluted with methanol to 50 ng/mL of NVP-d3 and 200 ng/mL of 2-OH NVP-d3. All of the above stock solutions were stored in the refrigerator.

Carotenoid Quantification in Plasma

The analyzed samples were −80°C stored K3-EDTA fasting plasma that had been drawn at baseline. Handling of all samples and standards was always done under cool conditions and avoiding exposure to light. Carotenoid standards: astaxanthin, canthaxanthin, E-β-apo-8′-carotenal, α-carotene, β-carotene, fucoxanthin, and lycopene were obtained from Sigma-Aldrich (St. Louis, MO, USA). Lutein was purchased from Cayman Chemical (Ann Arbor, MI, USA), zeaxanthin and β-cryptoxanthin were provided by Extrasynthese (Genay, Lyon, France). 13-Z-β-carotene and 9-Z-β-carotene were purchased from Carbosynth (Newbury, Berkshire, UK).
Methanol of LC-MS grade, n-hexane, ethanol, and methyl tert-butyl ether (MTBE) of HPLC grade, blank human plasma, and butylated hydroxytoluene (BHT) were obtained from Sigma-Aldrich. Ammonium acetate (AMAC) and acetic acid of HPLC grade were purchased from Panreac Quimica SLU (Barcelona, Spain). Ultrapure water (Milli-Q) was generated by a Millipore system (Bedford, MA, USA).

Carotenoid Analysis in Fasting Plasma
