We used the Th1/Th2 U-PLEX (K15069L-2; Meso Scale Discovery [MSD]) platform to analyze cytokines in protein extract and BAL supernatant. This platform uses a 10-spot U-PLEX plate and unique linkers to analyze samples. Biotinylated capture Abs are assigned to linkers, which correspond to unique spots in the U-PLEX plate. The analytes tested were IFN-γ, IL-1β, IL-2, IL-4, IL-5, KC, IL-10, IL-12p70, TNF-α, and IL-13. The MSD instrument applies a voltage to the plate electrodes, causing the capture labels to emit light. The intensity of the emitted light is measured. The BAL supernatant was at 1:1 dilution, and the protein extraction was at 1:2 dilution. Dilutions were used in the assay and then corrected for in the analysis. Blood was collected via cardiac puncture and stored in K2EDTA tubes.
