Quantifying Cellular Proliferation in Hypoxia

The proliferation ratio was determined in 3 biological replicates, with Cell Proliferation ELISA, BrdU, and colorimetric (#11 647 229 001, Roche, Basel, Switzerland), according to the manufacturer’s protocol. Briefly, 5000 cells were seeded in a 96-well plate in 100 µL/well and incubated at hypoxic conditions at 37 °C for 48 h. BrdU labeling was performed for 2 h. Cells were incubated with the anti-BrdU solution for 90 min and then with a substrate solution for 10 min. The absorbance was measured at 370 nm and calculated by subtracting the blank control absorbance value. Statistical results were calculated in GraphPad Prism6 (RRID:SCR_002798, GraphPad Software).
