Quantitative Western Blot Analysis

For Western blots, 2 × 106 K562 cells were treated with indicated reagents, lysed using radioimmunoprecipitation assay (RIPA) buffer and probed with indicated antibodies [31 (link)]. Twenty-five micrograms of protein from each lysate was run on a SDS/PAGE (10 %gel) followed by transfer to PVDF membrane. Membranes were incubated overnight at 4 °C with primary antibodies to either PPARα (1:250) or HIF-1α (1:250). Membranes were washed and incubated with the appropriate secondary antibodies (1:10000), followed by development using the Clarity chemiluminescent kit (Bio-Rad). The membranes were stripped and re-probed with an antibody for β-actin (1:20000), as a loading control. Quantification of images was performed using the ImageJ software suite.
