The desalted 5′-(6-aminohexyl)-modified Sgc8-c aptamer (Mw = 12.8 kDa, 5′-6-aminohexyl-ATC TAA CTG CTG CGC CGC CGG GAA AAT ACT GTA CGG TTA GA -3′, Sgc8-c-NH2) was purchased from IDT Technologies (Integrated DNA Technologies Inc., Coralville, IA, USA). The probe Sgc8-c-Alexa647 (Mw = 13.7 kDa, Figure S1A) was prepared and purified using Sgc8-c-NH2 and an activated-Alexa647 (Thermo Fisher Scientific Inc., Waltham, MA, USA), as previously it was reported by Calzada et al. [14 (link),45 (link)]. Finally, the probe was lyophilized without any additives and stored at −20 °C. Preformed pegylated liposomes (PPL) were gently gifted by MR-Pharma S.A. (Buenos Aires, Argentina). Micelle-forming copolymers—(i) poloxamer-based Pluronic® 127 (F127, Mw = 12.6 kDa, PEO 70% w/w, HLB 18), (ii) pH-dependent poloxamine-based Tetronic® 1307 (T1307, Mw = 18 kDa, PEO 70% w/w, HLB 27) and (iii) 908 (T908, Mw = 25 kDa, 80% w/w PEO, HLB 31)—were provided by BASF Corporation (New Milford, CT, USA, Figure S2A,B).

Pluronic and Tetronic-Based Liposomal Formulation

Pluronic® 127 (127; molecular weight: 12,600 g/mol; PEO content 70% by weight) and Tetronic® 1307 (T1307; molecular weight: 18,000 g/mol; PEO content 70% by weight) were supplied by BASF Corporation (New Mildorf, CT, USA) and used as received. Unilamellar preformed liposomes (LPS) composed by cholesterol (2 mg/mL), hydrogenated soy phosphatidylcholine (6 mg/mL) and phosphatidylethanolamine (4 mg/mL) grafted to a short chain of polyethylene glycol (Mw~2 kDa) were gently gifted by MR-Pharma S.A. (Buenos Aires, Argentina). Dimethylsulfoxide (DMSO, analytical quality) was purchased from Sigma-Aldrich (St. Louis, MO, USA). In all the cases, the water was purified and deionized (18 MΩ/cm2) in a Milli-Q water filtration system (Millipore Corp., Milford, CT, USA).

A generous donation of isosorbide dinitrate (ISDN, USP, 40 mg) was provided by the EIPICO Company in Cairo, Egypt, bioavailability of ISDN is approximately 25%. P-Toluene Sulphonic Acid, Maleic anhydride, Zinc chloride, Calcium chloride, zinc oxide, xylene and diethyl ether from Alpha Chemika (India) and magnesium oxide from BDH laboratory supplies in Poole, BH151TD (England). From BIO CHEM (Egypt), Polyvinyl alcohol (M.wt 10.000), decalin and chloroform (assay 99%), pure ethyl alcohol (assay 99.9%), Ferric oxide, Magnesium chloride, Arsen (III) oxide, Acetone, Magnesium metal, Phthalic anhydride, Tin oxide, Antimony oxide, potassium hydroxide, phenphethalin, and stannous chloride dihydrate. Calcium oxide and l(+) lactic acid (assay 88–92%) were obtained from LOBA CHEMIE (India). Aluminum oxide was purchased from RIEDEL–DE HAEN AG SEELZE-HANNOVER (China). Zinc metal, tween 80, anhydrous hydrogen phosphate, Potassium dihydrogen orthophosphate, and Sodium chloride were purchased from ADWIC (Egypt). Pluronic F88 and Tetronic 1307 were gifted from BASF, USA. Without additional purification, all substances were utilized precisely as they were given.

Synthesis of Gold Nanoparticles

All other reagents were analytical grade.
