The following are the lot numbers and purities of each compound: aprotinin (lot, 62009; purity of ≥98.0%), bromhexine hydrochloride (lot, 15159; purity of 99.39%), niclosamide (lot, 15718; purity of 98.68%), ciclesonide (lot, 0472344-2; purity of ≥98.0%), remdesivir (lot, 46182; purity of 99.78%), EIDD-2801 (lot, 67548; purity of 99.94%), and EIDD-1931 (lot, 0590872-1; purity of ≥95.0%). (Burlingame, CA, USA), respectively.

Antiviral Efficacy of 5-FU and EIDD-1931 against LASV

5’-fluorouracil (Cat# 03738, Sigma) and N4-hydroxycytidine (EIDD-1931) (Cat#9002958, Cayman) were dissolved in DMSO (cell culture grade, Sigma) and further diluted in DMEM media contain 2% FBS and 0.1% DMSO. Vero cells were mock-treated or treated with 5-FU or EIDD-1931 at different concentrations as indicated in each experiment for 4 hrs and then infected with wt rLASV and ExoN- rLASV (MOI 0.1) for 1 hr. Virus inoculums were removed and replaced with fresh media (DMEM + 2% FBS and 0.1% DMSO) containing 5-FU or EIDD-1931 of different concentrations as indicated. At 48 hpi, the supernatants of infected cells were harvested and subjected to plaque assay to determine virus titers. The viability of Vero cells treated with 5-FU and EIDD-1931 alone were assessed using CellTiter-Glo Viability Assay (Promega).
