Quantifying YAP/Lamin A/C Phosphorylation

Protein lysates from control/DMSO and VP (YAP inhibitor) were separated by electrophoresis on 4–12% bis-Tris SDS PAGE gels, with MOPS buffer and transferred to a nitrocellulose membrane. After blocking, the membranes were incubated overnight for 10 hours at 4°C with the primary antibodies: Rabbit Polyclonal anti-phospho YAP (ser127) (Cell Signaling, #4911S) at 1:1000 dilution or Rabbit Monoclonal anti-phospho Lamin A/C (Ser22) (D2B2E) (Cell Signaling, #13448S) at 1:1000, Rabbit Monoclonal anti-YAP (D8H1X) XP® (Cell Signaling, #14074S at 1:1000, Mouse Monoclonal anti-Lamin A/C (4C11) (Cell Signaling, #4777S) at 1:1000, and Rabbit Monoclonal anti-GAPDH (Cell Signaling, #2118S) at 1:1000. These primary antibodies were detected using Polyclonal Goat anti-Rabbit coupled to IRDye 680RD (LI-COR Bioscences, #925–68071) at 1:10000 or Polyclonal Goat anti-Mouse coupled to IRDye 680RD (LI-COR, #925–68070) at 1:10000.
