Confirming 3T3 IL-2/IL-21/CD40L Cell Line

Initial characterization of the 3T3 IL-2/IL-21/CD40L cell line was performed to confirm continued CD40L expression. CD40L expression on non-irradiated 3T3 IL-2/IL-21/CD40L cells was confirmed by flow cytometry after 1 week (3 passages) and 8 weeks (24 passages). Cells were detached from the flask using cold PBS supplemented with 5 mM EDTA (Gibco) and a portion of the sample was transferred to FACS tubes. Cells were incubated with anti-CD154 PE (BD Biosciences, TRAP-1) in stain media (1× PBS, 10% FBS) for 20 min at 4 °C. The sample was washed with PBS and run on an LSR II (BD Biosciences) alongside an unstained cell sample. Data were analyzed using FlowJo v9.4 (BD Biosciences).

Immunophenotypic Characterization of MSCs

Aliquots of 1 × 105 MSCs were washed and suspended in PBS supplemented with 0.1% FBS. Cells were incubated with anti-CD34-PE, anti-CD45-PE, anti-CD73-PE, anti-CD90-FITC, anti-CD105-FITC, anti-CD80-FITC, anti-CD86-PE, anti-CD154-PE, anti-CD40-PE, anti-HLA-ABC-FITC, or anti-HLA-DR-FITC antibodies (all from BD, San Diego, CA, USA) for 30 minutes at 4°C and then washed twice in PBS containing 0.1% FBS. Cells were then resuspended in 200 μL PBS containing 0.1% FBS and analyzed at 10,000 events per test using the FACSCalibur (BD Biosciences) or NovoCyte (ACEA Biosciences Inc., San Diego, CA, USA). Data were analyzed using NovoExpress 1.2.1 software.

Isolation and analysis of activated CD4+ T cells

PBMCs were isolated from patient blood samples by density gradient centrifugation (Ficoll-Paque Plus; GE Healthcare). Fresh PBMC were cultured in AIM V medium (Gibco) for 20 h with 100 μg/ml peanut protein extract. The peanut extract was prepared by agitating defatted peanut flour (Golden Peanut and Tree Nuts) with PBS, centrifugation, and sterile-filtering. anti-CD154-PE (clone TRAP1; BD Biosciences) was added to the cultures for the last 3 h. After harvesting, the cells were labeled with anti-CD3-AF700 (clone UCHT1), anti-CD4-APC-Cy7 (RPA-T4), anti-CD45RA-FITC (HI100), anti-CD154-PE (all from BD Biosciences), anti-CD69-AF647 (FN50; BioLegend), and Live/Dead Fixable Violet stain (L34955; Thermo Fisher). Live CD3+CD4+CD45RA activated CD154+ were sorted with a FACSAria II instrument (BD Biosciences). The sorted cells were processed via Seq-Well18 (link).
