Metabolic and Renal Biomarkers in Rats

Check if the same lab product or an alternative is used in the 5 most similar protocols
Blood glucose was measured in whole blood from rats with a Quick Auto Neo GLU-HK (SHINO-TEST CORPORATION, Tokyo, Japan). HbA1c was measured in whole blood from rats with a RAPIDIA Auto HbA1c-L system (FUJIREBIO Inc, Tokyo, Japan). Serum BUN and creatinine were measured by SRL Inc. (Wako, Saitama, Japan). Collection of 24-hour urine was done in metabolic cages. Urinary albumin and urinary creatinine were measured by SRL Inc. Urinary protein excretion was determined with a Bio-Rad protein assay kit (Bio-Rad Laboratories, Hercules, CA, USA). Urinary albumin excretion was expressed as the ratio of albumin to creatinine.

Insulin Resistance Assessment Protocol

Check if the same lab product or an alternative is used in the 5 most similar protocols
As part of the health checkup, plasma glucose and serum insulin concentrations were measured at both baseline and follow-up surveys. Plasma glucose concentrations were assayed enzymatically using Quick-auto-neo-GLU-HK (Shino-Test Corp., Tokyo, Japan). Serum insulin concentrations were measured using a chemiluminescence immunoassay method at an external laboratory (Mitsubishi Chemical Medience Corporation, Tokyo, Japan). We computed the homeostasis model assessment of insulin resistance (HOMA-IR) using the following formula: HOMA-IR ¼ (fasting serum insulin [mU/mL] Â fasting plasma glucose [mg/dL])/405 [29] . A higher HOMA-IR value indicates greater insulin resistance.

Biochemical Assessment of Rat Diabetes

Check if the same lab product or an alternative is used in the 5 most similar protocols
Blood glucose was measured in whole blood from rats with Quick Autoneo GLU-HK (Shino-Test Corp, Tokyo, Japan). Hemoglobin A1c (HbA1c) was measured in whole blood from rats with RAPIDIA Auto HbA1c-L (Fujirebio Inc., Tokyo, Japan). Serum UN and creatinine were measured by SRL Inc. (Wako, Saitama, Japan). Twenty-four-hour urine was collected in metabolic cages, and urinary albumin and urinary creatinine were also measured by SRL Inc. Urinary protein excretion was determined with a protein assay kit from Bio-Rad (Hercules, CA, USA). Urinary albumin excretion was expressed as albumin/creatinine ratio.

Check if the same lab product or an alternative is used in the 5 most similar protocols
As part of the health checkup, the plasma glucose concentration was enzymatically assayed using Quick-auto-neo-GLU-HK (Shino-Test Corp., Tokyo, Japan), and HbA1c levels were measured by latex agglutination immunoassay using the Determiner HbA1c kit (Kyowa Medex Co., Ltd., Tokyo, Japan) at an external laboratory (Kinki Kenko Kanri Center, Shiga, Japan).

Plasma Lipid and Glucose Analysis

Check if the same lab product or an alternative is used in the 5 most similar protocols
After administration of pitavastatin for 4 weeks in eNOS−/−ApoE−/− mice (Study 1) or for 2 weeks in eNOS−/−ApoE+/+ mice (Study 2), blood was drawn through an inferior vena cava puncture and collected into a tube containing a 1/10 volume of heparin (10 U/ml) after overnight fasting and then centrifuged at 2000 g for 20 minutes. Plasma was stored at −80°C until use. Plasma levels of glucose were determined by an enzymatic method with Quickauto Neo GLU-HK (Shino-Test Corporation, Japan) using a Hitachi 7180 Clinical Analyzer (Hitachi, Tokyo, Japan). Plasma levels of lipids were also analyzed by an enzymatic method with type Wako CHO • H (Wako Pure Chemical Industries. Ltd., Tokyo, Japan) using the same analyzer as that for glucose.

Fasting Serum Biochemical Analysis

Check if the same lab product or an alternative is used in the 5 most similar protocols
Blood was collected by cardiocentesis from isoflurane anesthetized mice at 11 Am after 15 hours’ fasting and used for serum preparation. Serum levels of glucose, immunoreactive insulin, triglycerides (TGs), total cholesterol (T-Cho), aspartate transaminase (AST), and alanine transaminase (ALT) were measured using the Quick-auto-neo-GLU-HK (Shino-Test), Insulin Rat enzyme-linked immunosorbent assay (ELISA) kit (catalog No. ERINS, RRID: AB_2848197, Thermo Fisher Scientific), L-type Wako TG M, L-type Wako CHO M, L-type Wako AST J2, and L-type Wako ALT J2 (Fujifilm-Wako), respectively.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
