All animal procedures were performed in accordance with the protocol approved by the Institutional Animal Care and Use Committee at Stanford University. Six to 10-week-old mice on a C57BL/6J background purchased from The Jackson Laboratory, Bar Harbor, ME, were utilized for all studies: wild-type C57BL/6J mice (JAX#00664), GFAP-Cre (JAX#12886), ChAT-Cre (JAX#6410), Rosa26 eGFP (JAX#4077), Rosa26 Confetti (JAX#13731), and Rosa26 TDT (JAX#7914).

Genetically Modified Mouse Strains

Wild-type C57BL/6J (CD45.2), Rosa26Lox-Stop-Lox-tdTomato (AI1433) and Rosa26Confetti 34 mice were purchased from The Jackson Laboratory (strain numbers: 000664, 007914 and 013731, respectively). Strains that were kindly provided to us: AicdaCreERT2 21 by J.-C. Weill and C.-A. Reynaud (Université Paris-Descartes), S1pr2CreERT2 BAC-transgenic 25 (link) by T. Kurosaki and T. Okada (U. Osaka, RIKEN-Yokohama), CD20-CreERT227 (link) by M. Shlomchik (U. Pittsburgh) and Prdm1flox/flox 35 (link) by J. Boss (Emory University). OT-II TCR transgenic (Y-chromosome)36 (link) CD45.1 mice were bred and maintained in our laboratory. All mice were held at the immunocore clean facility at the Rockefeller University under specific pathogen-free conditions. All mouse procedures were approved by the Rockefeller University’s Institutional Animal Care and Use Committee.

Characterization of Genetically Engineered Mice

Rosa26confetti (Stock 017492), NOD scid gamma (NSG, Stock 005557), NSG-EGFP (Stock 021937), Rosa26mT/mG (Stock 007676), R26-M2rtTA (Stock 006965), pTRE-H2BGFP (Stock 005104), Prrx1-cre (Stock 005584), Ocn-cre (Stock 019509), Pax7-cre (Stock 010530), Shh-cre (Stock 005622), Mpz-cre (Stock 017927), MIP-GFP (Ins1-EGFP, Stock #: 006864), floxed Stat3 (Stat3fl/fl) mice (Stock 016923) and UBC-GFP (Stock 004353) mice were purchased from Jackson Laboratories. Mfge8−/− (RBRC01726) mice30 (link) were purchased from RIKEN bioresource research center. Shn3 loxp mice31 (link) were reported previously. All mice were maintained on a C57BL6/J background throughout the study except for experiments involving 4T1.2 cells that were conducted in BALB/cJ mice. All animals were maintained in accordance with the NIH Guide for the Care and Use of Laboratory Animals and were handled according to protocols approved by the Weill Cornell Medical College institutional animal care and use committee (IACUC).

Rosa26 Confetti and Cre ER Mouse Breeding

Rosa26-Confetti and Rosa26-CreER were purchased from the Jackson Laboratories and bred at Emory University. All animals were housed in an Emory University Department of Animal Resources facility (Atlanta, GA) and used in accordance with an Institutional Animal Care and Use Committee–approved protocol.

Genetically Modified Mice for Confetti Lineage Tracing

PF4-Cre24 (link) and Rosa26-Confetti23 (link) were purchased from The Jackson Laboratory and maintained and cross-bred at our animal facility (stock no: 008535 and 017492). vWF-Cre mice were a gift of W. Aird.25 (link) Mhy9 fl/fl mice were a gift from Dr. Gachet.26 (link) All strains were backcrossed to and maintained on C57BL/6J-background. If not otherwise stated, animals of same sex and age were randomly assigned to experimental groups, and PF4-Cre negative control animals were used. We used Rosa26Confetti/+ animals expressing Cre for Online Supplementary Figure S1. The other experiments were performed with homozygous Rosa26Confetti/Confetti mice expressing Cre. Cre negative animals were used as controls. All procedures performed on mice were approved by the local legislation on the protection of animals (Regierung von Oberbayern, Munich).
