MDA MB231 and H460 (5× 105) cells were incubated with APC anti-human CD124
(IL-4Rα) and respective IgG isotype antibody (Biolegend, San Diego) as a control for 1 h
at room temperature. B16F10 cells were also incubated with APC anti-mouse CD124 (IL-4Rα)
and respective IgG isotype antibody (Biolegend). The level of IL-4R expression was measured by
flow cytometry (BD Bioscience, San Jose, CA, USA). Ten thousand events were collected for each
sample, and the percentage of APC anti-human CD124 bound to cells was calculated by comparison
with isotype control.
