Multiparametric Flow Cytometry of Mouse Spleen Cells

Approximately 1 × 106 spleen cells were collected, resuspended in 100 μl of PBS and incubated with 1.5 μl of PE-conjugated anti-mouse CD4, PE/Cy5-conjugated anti-mouse CD8a and FITC-conjugated anti-mouse CD3, PE-conjugated anti-mouse CD19 and FITC-conjugated anti-mouse CD80, or FITC-conjugated anti-mouse CD86 antibodies (all from BioLegend) in the dark on ice for 30 min. The cells were then washed and analyzed using a FACSCalibur flow cytometer and CellQuest software (BD Biosciences).

Lipopeptide-Stimulated Dendritic Cell Phagocytosis and Cross-Presentation

Bone marrow‐derived DC and splenocytes were stimulated with lipopeptides as previously described.9, 10 The phagocytosis assay was performed using a Phagocytosis Assay Kit (Cayman Chemical Company, Ann Arbor, MI, USA). Briefly, BMDC were stimulated with each lipopeptide for 24 hours and then cultured with Latex Beads‐Rabbit IgG‐FITC Complex for 6 hours. For the phagocytosis and cross‐presentation assays with BMTC, BMDC were cultured with BMTC (BMDC:BMTC ratio = 1:5) for 12 h (phagocytosis) or 24 hours (cross‐presentation) and then analyzed by flow cytometry. Flow cytometry and the ELISA were performed as previously described9, 10 using the following antibodies and kits: FITC‐conjugated anti‐mouse CD80; phycoerythrin (PE)‐conjugated anti‐mouse MHC class I molecule Kb bound to the peptide SIINFEKL (Kb‐SIINFEKL; BioLegend, San Diego, CA, USA); allophycocyanin (APC)‐conjugated anti‐CD11c (eBioscience, San Diego, CA, USA); and ELISA kits specific for IFN‐γ and IL‐12p40 (R&D Systems, Minneapolis, MN, USA).

Hepatitis B Vaccine Immunomodulation Evaluation

The hepatitis B vaccine (consisting of HBsAg and aluminium adjuvant) and HBsAg were a generous gift from the Dalian Hissen BioPharm Co., Ltd. (Dalian, China). Cordycepin was purchased from Beijing Century Bioko Bio-technology Co. (Beijing, China). The endotoxin levels of these reagents were <0.1 EU/ml as determined by an endotoxin detection assay (Tachypleus Amebocyte Lysate, Xiamen, China). Fluorescein isothiocyanate (FITC)-conjugated anti-mouse CD3, phycoerythrin (PE)-conjugated anti-mouse CD4, PE/Cy5-conjugated anti-mouse CD8a, FITC-conjugated anti-mouse CD80, FITC-conjugated anti-mouse CD86 and PE-conjugated anti-mouse CD19 antibodies were all purchased from BioLegend (BioLegend, San Diego, CA, USA).

Flow Cytometric Analysis of Dendritic Cell Markers

RAW264.7 cells were treated, harvested, washed with PBS, and stained with 1 μg phycoerythrin (PE)-conjugated anti-mouse CD86 antibodies and fluorescein isothiocyanate (FITC)-conjugated anti-mouse CD80 or PE-conjugated anti-mouse CD40 and FITC-conjugated anti-mouse I-AK antibodies (Biolegend) for 30 min at room temperature. After washing and resuspending with PBS, the cells were analyzed using a FACSCalibur machine (Becton Dickinson, USA). The data were processed using Cell-Quest data analysis software.

Synthesis and Characterization of PEG-Peptide Conjugate

20 kDa 4-arm PEG-azide was purchased from JenKem Technology USA (Beijing, China). Tris(3-hydroxypropyltriazolylmethyl)amine (THPTA) and sodium ascorbate (NaAsc) were purchased from Sigma-Aldrich (St. Louis, MO). Copper (II) sulfate pentahydrate (CuSO4 • 5 H2O) was purchased from Acros Organics (Geel, Belgium). Alkyne functionalized peptide bearing an N-terminal 4-pentynoic acid (homopropargyl, hp) modification, hpPLP139–151 (hp-HSLGKWLGHPDKF-OH), was obtained from Biomatik, USA, LLC (Wilmington, DE). All reagents were used as received without further purification. For in vitro cell assays and in vivo studies, female 4–6-week-old SJL/J (H-2) mice were purchased from Envigo Laboratories (Indianapolis, IN). For EAE induction, incomplete Freund’s adjuvant (IFA) and heat-killed mycobacterium tuberculosis H37RA were purchased from Difco (Sparks, MD). Additionally, pertussis toxin was purchased from List Biological Laboratories (Campbell, CA). For use in flow cytometry, TruStain fcX (anti-mouse CD16/32), R-phycoerythrin (PE)/Cy7-conjugated anti-mouse CD3, PE-conjugated anti-mouse CD86, FITC-conjugated anti-mouse CD80, AlexaFluor647-conjugated anti-mouse CD19, and BV421-conjugated anti-mouse CD11c were purchased from BioLegend (San Diego, CA).
