Cell culture medium and supplements were purchased from Life Technologies (Grand Island, NY, USA). Antibodies against phosphorylated AKT (Ser473) (p-AKT) (#4060S), AKT (#2920S), phosphorylated GSK3β (Ser9) (p-GSK3β) (#9336S), GSK3β (#9832S), and β-actin (#3700) were obtained from Cell Signaling Technology (Beverly, MA, USA), and antibody against OPA1 (#612606) was obtained from BD Biosciences (San Jose, CA, USA). Chamber slides, goat anti-rabbit (#656120) and anti-mouse (#626520) secondary antibodies, Lipofectamine 3000 (#L3000015), and 4′,6-diamidino-2-phenylindole (DAPI) (#P36931) were obtained from Invitrogen (Carlsbad, CA, USA). MitoSOX Red (#M36008), tetramethylrhodamine methyl ester (TMRM) (#T668), MitoTracker Green (MT Green) (#M7514), and MitoTracker Deep Red (#M22426) (all from Molecular Probes, USA) were purchased from Life Technologies. Terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) assay kits (#11684795910) were obtained from Roche (Mannheim, Germany). HT (#H4291), H2O2 (#88597), 4-benzyl-2-methyl-1,2,4-thiadiazolidine-3,5-dione (TDZD-8) (#T8325), LY294002 (#L9908), N-acetyl-l-cysteine (NAC) (#A7250), and 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) (#M2128) were obtained from Sigma-Aldrich (St. Louis, MO, USA). ATP assay kit (#S0026) was obtained from Beyotime Institute of Biotechnology (Shanghai, China).

Ovarian Cancer Cell Line SKOV3 Culture

Human ovarian carcinoma cell line SKOV3 was obtained from the American Type Culture Collection (Manassas, VA, USA) and maintained in McCoy’s 5A medium (Gibco®; Thermo Fisher Scientific, Waltham, MA, USA), supplemented with 10% fetal bovine serum (FBS), 100 U/mL penicillin, and 100 μg/mL streptomycin at 37°C and 5% CO2. Paclitaxel-resistant cell subline, SKOV3-TR30, was developed by exposing parental SKOV3 cells to increased concentration of paclitaxel as described previously.14 (link) Two GSK-3 inhibitors, lithium chloride (LiCl) and 4-benzyl-2-methyl-1,2,4-thiadiazolidine-3,5-dione (TDZD-8), were purchased from Sigma-Aldrich (St Louis, MO, USA).

Investigating Mitochondrial Dynamics in Diabetes

Drugs were prepared as stock solutions and were diluted to the final concentration immediately before use. Incubation of hippocampal slices with drugs was performed in either a recovery chamber or interface-recording chamber as needed. Drugs were added to culture plate wells. Final concentrations and sources of the drugs were as follows: 4-benzyl-2-methyl-1,2,4-thiadiazolidine-3,5-dione (TDZD8) (5 μmol/L; Sigma-Aldrich), mdivi-1 (10 μmol/L; Sigma-Aldrich), and glucose (50 mmol/L; Sigma-Aldrich). The final concentration of vehicle control DMSO was less than 0.5% in all experiments. For chronic in vivo administration of mdivi-1, the drug was dissolved in DMSO (10 or 25 mg/mL) as stock solution and further diluted in saline (DMSO: saline = 1:9) immediately before daily injection in diabetic or nondiabetic male mice at a dosage of 10 or 25 mg/kg i.p. Drug was administered once a day beginning at 19 weeks of age and continued for 2 weeks. The same amount of DMSO dilution in saline was prepared and then administered to age-matched male nondiabetic and diabetic littermates as controls. One day after the end of treatment, animals were subjected to either electrophysiological studies or biochemistry assays.
