Formulation and Characterization of Amoxicillin Trihydrate Hydrogel

Amoxicillin trihydrate (AMT) of analytical grade was obtained from Thermo Fisher Kandel GmbH (Karlsruhe, Germany), poloxamer (Kolliphor® P407) was purchased from BASF (Ludwigshafen, Germany). Bovine serum albumin (BSA, lyophilized powder, purity ≥ 97%),ethanol 96% v/v, mucin from porcine stomach (Type III), and all reagents were purchased from Merck Ltd. (Budapest, Hungary) if not indicated otherwise. Sodium hyaluronate (NaHA, Mw = 1400 kDa) was donated from Gedeon Richter Plc (Budapest, Hungary). Analytical-grade methanol was purchased from Molar Chemicals (Budapest, Hungary). Purified water was filtered using the Millipore Milli-Q® (Merck Ltd., Budapest, Hungary) Gradient Water Purification System. All other solvents and reagents used in this study were of pharmaceutical grade.

Poloxamer-Based Intranasal Ketamine Formulations

Example 1

18 g of poloxamer 407 (Kolliphor P407®, BASF) were dissolved under cold conditions, in 100 ml of water, then 9.23 g of ketamine hydrochloride (i.e. 8 g of ketamine) were added and dissolved while stirring for approximately one hour.

The solution (F1) obtained gels on contact with the mucous membranes of the buccal cavity.

The formulations F2 to F7 reported in table I below were prepared according to the same procedure. As regards the formulations F3 and F4, the mucoadhesive agent (alginate) was added beforehand to the solution of poloxamer before addition of the ketamine at a temperature between +40° C. and +70° C.

Poloxamer-Chitosan Ophthalmic Formulations

Poloxamers P407 (Kolliphor P407; EO 98 PO 69 EO 98 ; average molecular weight (M w ) 13,498 g/mol) and P188 (Lutrol micro F68; EO 80 PO 27 EO 80 ; average M w 8902 g/mol) were purchased from BASF SE (Ludwigshafen, Germany). More than 90% deacetyled ultrapure PROTASAN™ chitosan salt (Protasan UP CL 214; M w 150,000-400,000 g/mol; viscosity 20-200 mPa  s) (CS) was obtained from NovaMa-trix® (Sandvika, Norway). Timolol maleate (TIMO), dexamethasone (DEX), dorzolamide hydrochloride (DORZO) were kindly donated by TEVA Pharmaceutical Industries Ltd. Tobramycin (TOBRA) standard was purchased from the United States Pharmacopeia (Rockville, MD).
Simulated tear fluid (STF), pH 7.4, was prepared by dissolving the following substances in double-distilled water: KCl (1.4 mg/mL), NaCl (6.8 mg/mL), NaHCO 3 (2.2 mg/mL), and CaCl 2 Â 2H 2 O (0.08 mg/mL). 23 Final step in STF preparation was filtration of solution (Whatman™ membrane filters, 0.2 mm, RC58; GE Healthcare Limited, Buckinghamshire, UK).
Cellulose acetate 0.8 mm membranes for in vitro release (IVR) test (CA 0.8) were obtained from Sartorius Stedim Biotech (G€ ottingen, Germany).

Poloxamer-Based Antimicrobial Peptide Formulation

Poloxamer 407 (Kolliphor® P407; kindly provided by BASF, Ludwigshafen, Germany) and methyl paraben (Fluka, Buchs, Switzerland) were dissolved in 20 mM acetate buffer pH 5.5 in an ice bath for 1–2 h until the polymer was completely dissolved (23 wt% poloxamer 407 and 0.03 wt% methyl paraben). For formulations containing DPK-060 without a nanocarrier, DPK-060 was first dissolved in acetate buffer and then added to the polymer solution while stirring. The final concentrations of poloxamer 407 and methyl paraben were 17 wt% and 0.02 wt%, respectively; the final concentrations of DPK-060 were 0.25 wt%, 0.5 wt%, and 1.0 wt%, corresponding to approximately 1.0 mM, 2.0 mM, and 4.0 mM, respectively.

Zein-Based Drug Delivery System

The non-essential amino acid was purchased from Gibco (Paisley, UK). Amicon Ultra filter 100 kDa (Ultracel membrane with 100,000 MWCO, Millipore Corporation, Bedford, MA, USA). Polyethylene terephthalate membrane inserts were purchased from Millicel® Merck (Darmstadt, Germany). Ultrapure water was obtained using a Milli-Q purification system (Merck Millipore). All other solvents and chemicals were of analytical grade or equivalent.

Clopidogrel Solid Dispersions Using Hydrophilic Polymers

In this study, clopidogrel hydrogensulfate (Ph. Eur. 10.0) was used as a model API. Four different hydrophilic polymers: poloxamer 407 (Kolliphor® P407, BASF, Germany), macrogol 6000 (Polyglykol 6000 S, Clariant Export AG, Switzerland), povidone (Kollidon® 30, BASF, Germany), and copovidone (Kollidon® VA 64, BASF, Germany), were used as carriers. Ethanol 96% (v/v) was used as a solvent for SDs preparation by solvent evaporation method. All excipients were of pharmaceutical grade.
Other reagents and chemicals used for the characterization of SDs were of analytical grade. API, excipients, and other reagents and chemicals were kindly provided as a gift from Bosnalijek d.d. (Sarajevo, Bosnia and Herzegovina).
