Bioreactor Cell Culture Sampling

The bioreactor was sampled daily for off-line measurements. Total cell density, cell viability, and cell size were measured using a cell counter (Vi-cell Beckman Coulter, Fullerton, CA). Glucose, lactate, ammonium, glutamine, and other metabolites were measured with a NOVA Flex BioProfile analyzer (NOVA biomedical). pH was measured using Seven Excellence Multiparameter (METTLER TOLEDO). Oxygen (pO2) and carbon dioxide (pCO2) partial pressure were measured using a Siemens Blood Gas Analyzer RapidLab 248. Technical replicate samples of 15 mL were collected daily from pre-inoculation day to day 13, centrifuged at 2000 r.c.f. for 10 min, filtered through a 0.2 µm polyethersulfone filter (Pall), and stored at −80 °C in 500 µL aliquots in matrix tubes (Thermo Fisher) for further analysis. Technical replicate bulk sampling of 100 mL aliquots was performed from day 5 to day 13. On termination day (day 13), 2 L of the total working volume was collected and centrifuged at 2000 r.c.f. for 60 min, filtered through a 0.2 µm polyethersulfone filter (Pall), and stored at −80 °C.
