Fibroblast Cell Culture Protocol

Poly(vinyl alcohol) (PVA), Poly(ε-caprolactone) (PCL), Agar-Agar (powder BioScence–Grade, ROTH, Nuremberg, Germany), Standard agarose (powder, ROTH, Nuremberg, Germany), Fluorescein Isothiocyanate-Dextran (wt. 10,000, Sigma-Aldrich, St. Louis, MO, USA) (FSC), H3PO4, and Glyoxal (Sigma-Aldrich, MO, USA) were used. Mouse embryo fibroblast BALB/3T3 clone A31 cells (ATCC CCL-163) were cultured as adherent monolayers in plastic Petri dishes Nunc™ (Fisher Scientific, Waltham, MA, USA) in Dulbecco’s modified eagle medium (4 mM L-glutamine, 4500 mg/L glucose, 1 mM sodium pyruvate, and 1500 mg/L sodium bicarbonate, Sigma-Aldrich) supplemented with 10% (v/v) fetal bovine serum (Sigma-Aldrich, MO, USA) and antibiotic–antimycotic solution (Sigma-Aldrich, MO, USA). MilliQ water was used for the preparation of all aqueous solutions (Milli-Q® IQ 7005 Water Purification System, Millipore, Billerica, MA, USA). All reagents and chemicals used in experiments were of analytical grade and were used as received.
