VSMC Treatments and Morphological Changes

Aortas were obtained from Sprague-Dawley rats, and VSMCs were grown from aortic explants and used at passages 3 to 6. Cells were treated with different conditions when they reached 75–85% confluence: (1) AngII (Sigma) at 1 × 10−6 mol l−1 (determined by prestudy) for 48 h; (2) emodin (Pfizer, Groton, CT, USA) 10, 40 and 80 μmol l−1 for 48 h; (3) miR-126 mimic and Lipofectamine 2000 reagent for 6 h, then LiCl (20 mmol l−1) and/or AngII (1 × 10−6 mol l−1) for 48 h; and (4) miR-126 inhibitor and Lipofectamine 2000 reagent (Invitrogen, Milan, Italy) for 6 h, then AngII (1 × 10−6 mol l−1) for 48 h. The morphology of the cultured cells was observed by electron microscopy. Each treatment was performed at least 3 times.

Angiotensin II-Induced Hypertension Model

Procell Life Science and Technology Co. Ltd (Wuhan, China) provided hVSMCs. One percent penicillin-streptomycin solution (P/S) and 10% fetal bovine serum (FBS) were added to Dulbecco's modified eagle medium (DEME, Gibco, New York) for cell culture. Cells were incubated within the humid incubator under 37°C and 5% CO 2 conditions. For cell treatment, Ang II-induced hypertension was established after hVSMCs treated with 1 μM Ang II (Invitrogen, California, USA) at 0, 3, 6, 12, 24, and 48 h. The viability of 24 h hVSMCs was about 60%, which is suitable for follow-up functional research. Therefore, Ang II-induced hypertension model was established after hVSMCs treated with 1 μM Ang II for 24 h.

Reagents are as follows: RNA extraction and PCR reagents (Roche, Switzerland), TUNEL antibody (Abcam, USA), phosphatase inhibitor (Roche, Switzerland), atrial natriuretic peptide (ANP)/transforming growth factor-β (TGF-β)/Collagen III (Col III)/bcl-2/bax/caspase-3/ERK/p-ERK/p38MAPK/p-p38MAPK/JNK/p-JNK/β-actin antibodies (Cell Signaling Technology, USA), BCA quantitation and Western Blot reagents (Invitrogen, USA), and PCR primers and reagents (Invitrogen, USA).
