PLX4720 (>99%, molecular weight 413.83 g/mol) was obtained from Selleck Chemicals (Houston, TX, USA). Water (ULC–MS grade), methanol (HPLC grade) and acetonitrile (HPLC‐S grade) were obtained from Biosolve (Valkenswaard, The Netherlands). Formic acid of analytical grade was supplied by Merck (Darmstadt, Germany). Water, purified by reversed osmosis was available at the research institute and used for all other purposes. Both pooled blank human lithium–heparin plasma and pooled female mouse lithium–heparin plasma were provided by Sera Laboratories (Haywards Heath, UK). Blank mouse tissue homogenates were prepared by mixing the whole organ with ice‐cold 2% (w/v) bovine serum albumin in Milli‐Q water followed by homogenization using the FastPrep‐24™ 5G instrument (M.P. Biomedicals, Santa Ana, CA, USA) for 1 min. Different appropriate volumes were used for different organs (1 ml for brain, spleen and lung; 2 ml for kidney; and 3 ml for liver and SIWC). All matrices for analysis were stored at −30°C.

Bile Acid Preparation and Analysis

Quantification of EGFR Inhibitors in Plasma

Erlotinib (as hydrochloric acid) and encorafenib were supplied by Sequoia Research Products (Pangbourne, UK). Afatinib, 13 C 6 -afatinib, 13 C 6 -erlotinib (as hydrochloric acid), OSI-420 (as hydrochloric acid, 95.9%), 13 C 6 -OSI-420 (as hydrochloric acid, 98.0%), gefitinib, gefitinib-d 8 and O-desmethyl-gefitinib were obtained from Alsachim (Strasbourg, France). Pooled human lithium-heparin plasma was supplied by Seralab Laboratories (Haywards Heath, West Sussex, UK), individual drug-free human plasma and pooled whole blood were obtained from Innovative Research Inc. (Novi, MI, USA).
