CCK-8 Assay for Cell Viability

Check if the same lab product or an alternative is used in the 5 most similar protocols
The CCK-8 assays were performed according to the manufacturer’s instructions (DOJINDO Laboratories, Kumamoto, Japan). Briefly, the cells were seeded in 96-well plates at a density of 1 × 104 cells/well, and incubated with different doses (0, 0.5, 1, 2, 4, 8 mM) of VC (Alfa Aesar, Ward Hill, MA,United States) for 3, 6, and 9 h. Then, 100 μl CCK-8 solution was added to each well. After that the cells were incubated for an additional 1–2 h. Finally, the light absorbance values were recorded at 450 nm using a Multiskan FC apparatus (Thermo Fisher Scientific, Waltham, MA, United States) and cell viability was calculated.

Evaluating CRNDE's Impact on HCT-116 Cells

Check if the same lab product or an alternative is used in the 5 most similar protocols
The effect of CRNDE on the proliferation and cisplatin sensitivity of HCT-116 cells was investigated using the CCK-8 assay. Harvested HCT-116 cells were seeded into 96-well plates at a density of 5×103 cells/well and cultured overnight at 37°C with 5% CO2. After treatment, the cells were incubated with 10 ml of CCK-8 solution (Bioswamp; Wuhan Bienle Biotechnology Co., Ltd.) for 4 h at 37°C with 5% CO2. The optical density of wells was measured at 450 nm using a Multiskan FC apparatus (Thermo Fisher Scientific, Inc.).

MTT Cytotoxicity Assay Protocol

Check if the same lab product or an alternative is used in the 5 most similar protocols
Ncm460 and HCT-116 cells were seeded into a 96-well plate (5 × 103 cells/well) and maintained at 37°C with 5% CO2 until the cells adhered. After treatment, 20 μl of MTT at a concentration of 5 mg/ml was added to each well for 4 h, and 150 μl of dimethyl sulfoxide (Sigma, MO, USA) was then added to each well. The plate was shaken for 10 min, and the absorbance was evaluated using a Multiskan FC apparatus (Thermo, Waltham, MA, USA) at 490 nm.

ELISA for Mouse Immunoglobulin Isotypes

Check if the same lab product or an alternative is used in the 5 most similar protocols
Maxisorp plates were coated using one of the following Abs: anti-IgG1, anti-IgG2a, anti-IgG2b, anti-IgG3, anti-IgM (Southern Biotech). After incubation with mouse sera, revelation was performed using biotin-coupled anti-mouse immunoglobulin (Ig) (H + L) (Jackson IR), streptavidin HRP (ThermoFisher), TMB, and H2SO4. Optical densities were assessed using a ThermoScientific Multiskan FC apparatus.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
