Generation and Validation of TM4SF5 Transgenic Mice

TM4SF5 transgenic C57BL/6 mice were generated (Macrogen, Seoul, Korea), and TM4SF5 expression was confirmed (37 (link)). The pcDNA3-hTM4SF5-FLAG plasmid (digested with NruI and DraIII), consisting of the cytomegalovirus promoter, whole human TM4SF5-FLAG sequence, and bovine growth hormone polyadenylation region, was used for the generation of C57BL/6 mice according to standard procedures, as explained previously (37 (link)). ApcMin/+ mice were kindly gifted by Dr. Young-Yun Kong (Department of Biological Sciences, Seoul National University, Seoul, Republic of Korea). ApcMin/+ mice were interbred with TgTM4SF5 to generate ApcMin/+:TgTM4SF5 offspring. All animal experiments were performed in accordance with the guidelines of the Seoul National University Institutional Animal Care and Use Committee (SNU-130225-9-2).

Bleomycin-induced Pulmonary Fibrosis Model

Wild-type C57BL/6 mice (4 weeks old) were purchased from Orient Bio Inc. (Seongnam, South Korea). Global TM4SF5-knockout C57BL/6 mice (Tm4sf5−/−) were generated by Macrogen (Seoul, South Korea). Briefly, base pairs of genomic TM4SF5 surrounding exon 1 were deleted using the RGEN/Cas9 genetic scissor. The genotypes of both wild-type and Tm4sf5−/− mice were determined by PCR analysis of genomic DNA obtained from tail clippings from 3-week-old animals. Male wild-type and Tm4sf5−/− mice at 8 weeks old were treated with either saline vehicle for the control or bleomycin (1 mg/kg, Santa Cruz Biotechnology) by intratracheal injection on day 0. Mice were sacrificed on day 28 for histological analysis and hydroxyproline assay.
