Cleaned up pooled single-cell libraries were analyzed using the Aglient Bioanalyzer 2100 HS kit. Libraries were sequenced at UBC Biomedical Research Centre (BRC) in Vancouver, British Columbia on the Illumina NextSeq 550 (mid- or high-output, paired-end 150-bp reads), or at the GSC on Illumina HiSeq2500 (paired-end 125-bp reads) and Illumina HiSeqX (paired-end 150-bp reads).

Single-Cell RNA-seq Library Preparation

Check if the same lab product or an alternative is used in the 5 most similar protocols
For each triplicate condition, a total of 1000 cells were dispensed in single wells and processed using the Smart-seq2 protocol96 (link). The amount of PCR cycles for the second-strand generation step were adjusted to 12 cycles to account for the increased input material. The unfragmented libraries were quality-controlled using a Bioanalyzer 2100 HS kit (Agilent). The libraries were then processed for tagmentation, using 1 ng of amplified cDNA as input, and 8 cycles were used for the dual-indexing PCR. The DNA molarity of the libraries after AMPureXP (Beckman Coulter) clean-up was assessed using a Bioanalyzer 2100 HS and a Qubit HS (Thermo Fischer) to estimate size distribution and DNA concentration respectively. Finally, the libraries were pooled at equimolar ratios and sequenced using a MiSeq v2 300 cycle kit (Illumina) (DNA Sequencing Facility, Dept. of Biochemistry).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
