Plasma Cytokine Quantification by Electrochemiluminescence

Electrochemiluminescence-based immunoassay was used to measure cytokine levels in plasma. Plates were then analyzed by the Meso Scale Discovery® (MSD) QuickPlex SQ 120 and samples concentrations were calculated using the Discovery Workbench 4.0 using a 4-PL curve fit model. Samples below the lower level of detection, which was calculated by the workbench software as 2.5 standard deviations above the assay background blank, were reported as 0 pg/mL.

Cytokine Profiling of LPS-Stimulated Cells

Samples were treated with 100 ng/ml lipopolysaccharide (LPS) for 24 h and assayed for human chemokine and cytokine biomarkers with V‐PLEX Human Cytokine 30‐Plex Kit (Meso Scale Discovery) following the manufacturer's instructions. Elution and Cytokine Analysis of NELF

The Leukosorb strips were eluted after thawing. The full protocol has been described elsewhere (Rebuli et al., 2017 (link)). Briefly, an elution buffer (1% bovine serum albumin [Sigma], 0.05% Triton X‐100 [Sigma], in DPBS [Sigma]) was added to the strip, then centrifuged to remove NELF from strips. NELF was then analyzed using a V‐Plex Human Cytokine 30‐Plex Kit (Meso Scale Diagnostics), according to manufacturer instructions and the minimum manufacturer recommended dilutions.

Cytokine Profiling of Activated iMGL

iMGL culture media was replaced with basal media for 2 hours prior to stimulation with IFNγ (20 ng/ml), IL1β (20 ng/ml), and LPS (100 ng/ml) for 24 hours, after which cells were collected for RNA and conditioned media assessed for cytokine secretion.
