Colon Motor Activity Analysis in Mice

Adult male NL3R451C and WT mice (25–35 g; 8–12 weeks of age) were euthanized by cervical dislocation. The entire colon (~5 cm in length) was dissected, flushed, and cannulated in an organ bath superfused with physiological saline (at 37°C, 95%O2/5% CO2, flow: 5 mL min−1). After a 30 min equilibration, video recordings were acquired using a Logitech Quickcam pro 9000 camera (30 fps, 640 × 840 resolution) in order to measure contractile motor activity for a duration of 3 hr. Motor activity was recorded at baseline (1 hr), with bath‐applied antagonist (1 hr), and during washout (1 hr). Pseudocolored spatiotemporal maps of motor patterns were created and motor pattern frequencies were quantified using in‐house software [Neal et al., 2009; Swaminathan et al., 2016] (Fig. S1). Bicuculline (10 μM, Tocris), gabazine (10 μM, Sigma), and CGP 54626 (100 nM, Tocris) were diluted in saline.

Optogenetic Manipulation of Striatal iMSN

We used optical stimulation to activate iMSNs selectively. Channelrhodopsin-2 (ChR2) and its reporter gene enhanced yellow fluorescent protein (EYFP) were inserted in a double-floxed inverted open reading frame viral vector and stereotaxically injected into the striatum of 1 month-old WT and R6/2 mice crossed with A2A-Cre mice (1.0 mm anterior and 2.0 mm lateral to Bregma, at a depth of 3.1 mm from the dura). After Cre recombination, ChR2-EYFP was selectively expressed in A2A-Cre neurons. To examine evoked GABA responses in GPe neurons the following protocol was used: a slow ramp voltage command (3 s duration) from a holding voltage of −70 mV to +10 mV was first applied. Then the cell was held at the new voltage for 2 s, after which the optical stimulation (470 nm, 1 mW, 0.5 ms) was delivered. After 2 additional s at +10 mV the membrane potential was brought back to the original holding potential. This protocol was repeated 3 times (interval between sweeps was 30 s) and the responses to light stimulation were averaged. SNAP5114 (GAT-3 antagonist, 20 μM), NO711 (GAT-1 antagonist, 10μM), or CGP 54626 (GABAB receptor antagonist, 1μM) (all from Tocris, Minneapolis, MN) were bath applied (10-20 min) to examine alterations in decay time (90%-10%) of the evoked GABA response measured with Clampfit v10.7 (Molecular Devices, Sunnyvale, CA; RRID:SCR_011323).

Thalamic Network Activity Modulation by AMPK Agonists

Network activity was recorded in 400 µm thalamic slices within an interface chamber supplied with warm, oxygenated artificial CSF (31–33°C, 95% O2/5% CO2). Electrical stimuli were delivered to reticular thalamus with bipolar tungsten electrodes (FHC), while single tungsten electrodes placed in ventrobasal nucleus recorded extracellular activity. Filtered activity (0.1–3 kHz) was digitized and recorded with a Digidata 1440A and pClamp software. AMPK agonists and CGP-54626 (Tocris, Cat. no. 1088) were dissolved in the artificial CSF perfusate. Activity was quantified with custom MATLAB scripts.36 (link) Voltage amplitude thresholds were applied to detect action potentials and were set at 3–4× the root mean square of the trace before stimulation. Oscillation duration was defined as the period between the stimulus onset and the last activity burst to occur within the specified inter-burst interval of 1 s.

Preparation of Common Pharmacological Agents
